摘  要:目的探究石决明水提液对白内障大鼠晶状体的保护作用机制。方法实验研究。于2012年12月至2013年12月在温州医科大学药学院实验室,将100只大鼠按体重编号采用随机数字表随机分为4组,每组25只,制作D-半乳糖性白内障大鼠模型后,空白组和模型组分别点生理盐水,白内停组给白内停滴眼液滴眼,石决明组给石决明水提液。造模成功后处死大鼠,取出晶状体,免疫组化测定CHOP表达,Western Blot方法检测GRP78的表达,荧光定量PCR测定XBP—1mRNA表达。结果石决明组和白内停组CHOP,GRP78,XBP—1mRNA的表达均低于模型组,有显著性差异(P〈0.01),石决明组CHOP和GRP78的表达与白内停组无显著性差异(P〉0.05),石决明组XBP—1mRNA的表达低于白内停组,有显著性差异(P〈0.01)。结论石决明水提液对白内障大鼠的晶状体有保护作用,作用等同或者优于白内停。Objective To investigate the protect mechanism of Shijueming on the rats with D-galactose cataract. Methods One hundred healthy SD rats were randomly divided into four groups (blank group, control group, Baineiting eyedrops group, Shijueming eyedrops group), D-galactose cataract models were established. Blank and control group were treated with physiological saline eye- drops. Baineiting eyedrops group was treated with Baineiting eyedrops. Shijueming eyedrops group was treated with 0.25% Shijueming eyedrops (three times each day, one drop each time). The levels of CHOP, GRP78 and XBP-1mRNA were measured by immunohistochemistry, Western blot and RT-PCR methods. Results The expression of CHOP, GRP78 and XBP-1mRNA in shijueming eye- drops group were lower than in control group (P 〈0.05). Conclusions The water extract of Shijueming can decrease endoplasmic reticulum stress in lens epithelial cells of rats with D-galactose cataract and protect lens.
