摘 要:为探讨小G蛋白Ran对β-酪蛋白合成的主要信号通路mTOR和Jak2/Stat5及其增殖的影响,本研究对Ran进行瞬时转染和稳定转染,Western blot检测细胞中mTOR、p-mTOR、Stat5和p-Stat5的表达;高压液相层析仪检测酪蛋白的分泌量;CASY细胞活力分析仪分析细胞活力。结果显示,Ran过表达时,细胞活力显著增加(P<0.01),细胞中mTOR、p-mTOR、Stat5和p-Stat5的表达均也显著增加(P<0.01);表明Ran可能激活细胞中的mTOR通路和Jak2/Stat5通路,对乳腺上皮细胞的泌乳与增殖有正向调节作用。Ran,a small G protein,belongs to a small GTPase superfamily,which is involved in various signaling pathways in cells,regulating and controlling cell cycles and protein synthesis.However,the knowledge about the impact of Ran on the major signaling pathways,mTOR and Jak2/Stat5 pathway leading to cell viability proliferation and milk protein synthesis is limited.Western blot were used to determine the mTOR,p-mTOR,Stat5,and p-Stat5 expression after Ran timely and stable transfection,theβ-casein secreted by BMECs was detected by high pressure liquid chromatography and the changes of the cell vitality was analyzed by using CASY Cell Counter.The results showed that after Ran was overexpressed,the cell viability was significantly increased(P〈0.01),furthermore the expression of mTOR,p-mTOR,Stat5,and p-Stat5 were also significantly increased(P〈0.01).These results reveal that Ran positively regulates Jak2/Stat5 pathway and mTOR pathway,leading to milk protein synthesis of BMECs.
