摘  要:目的:评价用于阿尔茨海默病研究的脑内注射β-淀粉样蛋白致痴呆动物模型的有效性及中医内涵。方法:通过对文献的回顾,从表面效度、预测效度以及结构效度三方面评价该模型的有效性。结果:该模型的表面效度成立,该模型可以模拟阿尔茨海默病样记忆障碍;预测效度成立,能够改善痴呆患者学习记忆功能的乙酰胆碱酯酶抑制剂可以改善该模型的学习记忆功能;结构效度成立,该模型的病理和生化变化符合淀粉样蛋白使神经元凋亡,从而导致痴呆发生的假说。结论:β-淀粉样蛋白具有中医内生毒邪的特点,脑内注射β-淀粉样蛋白致痴呆动物模型是用于研究阿尔茨海默病与内生毒邪相关的中医发病机制及治疗的有效工具。Objective: To evaluate the validity of dementia animal model established by intracerebral injecting with amyloid 13 -protein and explore its connotation of traditional Chinese medicine. Methods: To evaluate the effectiveness of dementia animal model from three aspects as face validity, predictive validity and construct validity based on the literature review. Results: The face validity of this animal model was true, because this animal model could be used to simulate the memory disorder of alzheimer disease. The predictive validity of this animal model was true, and its learning and memorial function could be improved by acetycholinesterase inhibitor which could improve the learning and memorial function of patients with alzheimer disease. The construct validity of this animal model was also true, in that the pathological and biochemical changes of this animal model were accorded with a hypothesis that the neuronal apoptosis caused by amyloid 13 -protein could lead to dementia. Conclusion: Amyloid 13 -protein had the characteristics of endogeny toxin pathogens of traditional Chinese medicine (TCM), and the dementia animal model established by intracerebral injecting with arnyloid 13 -protein was an effective tool used in treatments and researches on TCM mechanism related with alzheimer disease and endogeny toxin pathogens.
