桦菌芝多糖GF对S180荷瘤小鼠抑瘤作用及细胞超微结构的影响

摘  要:目的:研究桦菌芝多糖G-F组对荷瘤小鼠(S180)的抑瘤作用及对肿瘤组织细胞超微结构的影响,并探讨其抗肿瘤机制。方法:建立S180小鼠动物模型,随机分为对照组、桦菌芝多糖G-F低剂量组、桦菌芝多糖G-F中剂量组、桦菌芝多糖G-F高剂量组、猪苓多糖组。连续给药12 d后,利用颈部脱臼方法处死小鼠,将剥离的各组肿瘤组织分别称重,计算其抑瘤率;通过透射电镜观察S180荷瘤小鼠肿瘤组织细胞超微结构变化。结果:桦菌芝多糖G-F高、中剂量组抑瘤率分别为41.84%、41.38%,与模型对照组比较(P<0.01),具有显著性差异;通过透射电镜形态学观察显示,桦菌芝多糖能明显改善小鼠肿瘤组织细胞的病理形态学变化,诱导肿瘤细胞凋亡。结论:桦菌芝多糖G-F组具有抑制S180荷瘤小鼠肿瘤生长和诱导细胞凋亡的作用。Objective : To explore the mechanism of its anti-tumor, research on effect of pyropolyporus fomentaries sporophore polysaccharide GF content on anti- tumor and cell uhrastructure in S 180 Bearing- Mice. Methods: Establish S180 mice animal model, the mice randomly divided into the model control group, grifola polysaccharide group and modeling group and PFSPGF high, middle, and low-dose groups, check tumor tissues weighing, calculation of inhibition rate, carried out by electron microscopy observation of tumor tissues ultrastructure. Results : The inhibition rate of pyropolypous fomenarius sporophore Polysaccharides on the S180 tumor-bearing mice show that the high, middle and low dose group compared with model control group inhibit tumor growth of the S180 tumor mice in varying degrees, the middle and high dose group compared with the model control group ( P〈0.01 ) are obviously lower; the inhibition rate are 41.84%,41.38%; through the ceil morphology, pyropolypous fomenarius sporophore polysaecharides GF can promote tumor cell apoptosis and necrosis. Conclusions : Pyropolypous fomenarius sporophore Polysaccharides GF has a clear anti-tumor effect on the s180 tumor-bearing mice, promote tumor cell apoptosis and necrosis.
