摘 要:目的:研究ADAM10蛋白表达与口腔鳞状细胞癌淋巴结转移的相关性,并探讨其促进肿瘤细胞转移的可能机制。方法:采用免疫组织化学方法检测ADAM10蛋白在58例口腔鳞状细胞癌中的表达情况,并分析其与肿瘤淋巴结转移发生的相关性;采用Transwell法检测高、低转移潜能的HN-12和HN-4口腔鳞状细胞癌细胞系的迁移能力,并通过实时定量PCR法和蛋白质免疫印迹实验分别检测2株细胞中ADAM10基因和蛋白的表达水平;采用RNA干扰技术,沉默高转移潜能细胞HN-12中的ADAM10的表达水平,观察其对细胞迁移能力的影响。采用SPSS 16.0软件包对实验数据进行统计学分析。结果:ADAM10蛋白的高表达与口腔鳞状细胞癌发生淋巴结转移相关;在高转移潜能细胞系HN-12细胞中,ADAM10基因及蛋白表达水平较低转移潜能细胞系HN-4显著增高(P<0.01);HN-12细胞中沉默表达ADAM10基因后,细胞迁移能力显著降低(P<0.01)。结论:ADAM10蛋白的高表达与口腔鳞状细胞癌淋巴结转移相关;ADAM10促进肿瘤细胞的迁移能力是其影响肿瘤转移的可能机制。ADAM10有望成为治疗口腔鳞状细胞癌转移的新靶点。PURPOSE: To study the correlation of increased ADAM10 with lymph node metastasis in patients with oral squamous cell carcinoma and discuss the related mechanism of tumor metastasis. METHODS: Immunohistology was used to detect ADAM10 expression in oral squamous cell carcinoma tissues of 58 patients and the correlation of ADAM10 expression with lymph node metastasis was analyzed. Transwell assay was used to detect the migration ability of HN-12 cell, which is more metastatic, and HN-4 cell, which is less metastatic. Real-time PCR and Western blot were used respectively to detect the gene expression and protein expression levels of ADAM10 in these two cell lines. RNA interference technology was used to silence the ADAM10 expression level of HN-12 cell, and the effect on migration ability was observed. SPSS 16.0 software package was used for statistical analysis. RESULTS: Lymph node metastasis of oral squamous cell carcinoma was related to the high expression level of ADAM10; the expression of ADAM10 was much higher in HN-12 cell, which was more metastatic than in HN-4 cell(P〈0.01). Migration ability decreased after silencing the ADAM10 expression in HN-12 cell(P〈0.01). CONCLUSION: The high expression of ADAM10 was related to lymph node metastasis of oral squamous cell carcinoma. ADAM10 protein may promote tumor metastasis by enhancing cell migration. ADAM10 may be a new target in oral squamous cell carcinoma therapy.
