窖蛋白1在急性髓系白血病细胞中表达的初步观察

摘　　要：目的明确窖蛋白1(Caveolin-1,CAV1)在急性髓系白血病(AML)细胞中的表达及临床意义。方法收集AML患者骨髓标本83例、对照组11例,运用实时定量PCR(q PCR)方法检测CAV1基因m RNA的表达水平,分析CAV1在AML不同阶段表达特征及临床意义。结果⑴CAV1基因m RNA在AML初治组表达比对照组显著降低(P<0.05),初治组与缓解组表达无显著差异(P=0.123);难治组与复发组CAV1表达水平比初治组、缓解组、对照组显著升高(P<0.05)。⑵初治组中化疗缓解组与化疗未缓解组之间CAV1表达水平无显著差异。⑶AML初治组中CAV1基因m RNA表达水平与患者骨髓单个核细胞比例、外周血原始细胞比例、外周血白细胞计数无明显相关性(P>0.05)。结论 CAV1在AML中表达与白血病阶段有关,呈双向表达模式,随着病情进展而升高,可能成为预测白血病病情进展的指标之一。Objective To identify the expression pattern and clinical significance of Caveolin-1 （CAV1） mRNA in human a-cute myeloid leukemia （AML） cells. Methods Total 83 cases of Bone marrow samples were collected from AML patients,11 cases of non-neoplastic patients were used as controls. The expression level of CAV1 was detected by real-time quantitative PCR. Re-sults The CAV1 expression level in the new diagnosed group and remission group was significantly lower than the control group （P＆lt;0.05）. Both the relapse group and the refractory group had a significantly higher levels of CAV1 expression than the control group,the new diagnosed group and the remission group. The expression level of CAV1 in the new diagnosed AML patients which gain CR after chemotherapy had no significantly difference than AML patient who had not gain CR after chemotherapy. In new di-agnosed AML group,the expression level of CAV1 had no significantly correlation with the percentage of blasts cells in bone mar-row,the percentage of peripheral blood immature cells and the counts of white blood cell in peripheral blood. Conclusion CAV1 expression levels were various according to the stage of AML,and may be a factor to predict the progress of AML.
