摘 要:好氧反硝化菌在生物脱氮中有很好的应用前景,文章旨在从环境中分离具有高效脱氮能力的反硝化菌,对其进行分类及反硝化特性研究。以对硝酸盐氮的去除能力为主要指标,从对虾养殖池底泥分离筛选出有效去除硝酸盐氮的好氧反硝化菌,通过生理生化特性及16S r DNA序列分析对菌株进行初步鉴定。在好氧条件下,研究不同碳源、氮源、温度、p H值对该菌总氮去除率的影响。试验结果表明菌株S4在以柠檬酸钠为唯一碳源,30℃,100 r/min振荡培养24 h和36 h,硝酸盐氮去除率分别达到60%和72%。其16S r DNA序列与海源菌的同源性达98%,结合生理生化特点,初步确定菌株S4属于海源菌属(Idiomarina sp)。最适碳源、氮源、分别为乙酸钠和牛肉膏,最适培养温度为20℃,最适p H值为8,在最适条件下总氮去除率达80%。Aerobic denitrifiers have a good application prospect in the field of biological nitrogen removal process. The present study aimed at the isolation of aerobic denitrifiers with high nitrogen removing ability from environment and its phylogeny and denitfifying characteristics. Based on NO3--N removal, a strain was isolated from sediment of shrimp pond and identified according to its physiological and biochemical properties and phylogenetic analysis of its 16S rRNA sequence. The effects of culture conditions such as carbon source, nitrogen source, temperature and initial pH of medium were investigated under aerobic condition. Results showed that NOz--N removal rate of the strain S4 are respectively 60% and 72% under the conditions of sodium citrate as carbon source,temperature 30℃, shaking speed 100 r/min, respective for 24 h and 36 h culture. According to morphological characteristics, physiological and biochemical properties and sequence analysis of 16S rDNA, S4 is identified as Idiomarina sp.The most optimum carbon source and nitrogen source are respectively glucose and beef extract, and the most optimum temperature and pH value are at 20 ℃ and 8.0 respectively.
