摘 要:目的:评价新型控释性可注射牙槽骨修复材料在实验动物牙周缺损区缓释四环素的性能。方法:选用15只雄性健康杂交犬,在双侧上颌尖牙近中牙槽骨处制作三壁骨内袋缺损,分别随机将含1%、5%、10%四环素(TTC)的不饱和聚磷酸酯/β-磷酸三钙复合物(UPPE/β-TCP)直接注射入其中一侧的牙槽骨缺损区内(UPPE/β-TCP/TTC(1%)组、UPPE/β-TCP/TTC(5%)组和UPPE/β-TCP/TTC(10%)组)。于术后第3、7、14 d用滤纸提取术区的龈沟液,使用高效液相色谱法测定各组TTC的含量,对实验结果进行单因素方差分析和组间多重比较。结果:在第3、7、14 d时,三组的龈沟液TTC浓度均高于抑制牙周主要致病菌所需要的TTC最低抑菌浓度,在第7 d和第14 d时各组材料缓释四环素量的差异无统计学意义(P>0.05)。结论:含1%TTC的新型控释性可注射牙槽骨修复材料UPPE/β-TCP复合物在实验动物体内有较好的药物缓释性能,能够有效地抑制牙周致病菌,防止牙周炎的复发。Objective:The purpose of this study was to assess the in vivo release of drug in the periodontal pockets from the unsaturated polyphosphoester/β-tricalcium phosphate (UPPE/β-TCP) composite containing 1 %,5%or 10 %tetracycline (TTC) with experimental periodontitis in dogs. Method:Experimental periodontitis was induced by placing stainless-steel mesh on the mesial side of maxillary canines for 6 weeks in fifteen adult,healthy dogs. Subsequently, intrabony defects were resized so as to be standard,and UPPE/β-TCP composite containing 1%,5%and 10%TTC were directly injected in the experimental bone defects. The retention time of the antibiotic product was determined indirectly by measuring the concentration of TTC in gingival crevicular fluid (GCF) using high-performance liquid chromatography at 3,7 and 14 day after the placement of the UPPE/β-TCP/TTC composites. Result:The TTC concentrations in the GCF well above the minimum inhibitory concentration of most periodontal pathogens were seen at 3,7 and 14 day following the injection of the three UPPE/β-TCP/TTC composite formulations. Conclusion:The UPPE/β-TCP composite containing 1 % TTC formulations can be retained in the periodontal pocket for a prolonged period,releasing a sufficient amount of tetracycline to eliminate pathogenic bacteria.The local application of injectable bioerodible UPPE/β-TCP/TTC composites appears to be promising in the context of periodontal treatment.
