摘  要:对丙酮粉法提取金银花多酚氧化酶的工艺进行研究,结果表明,缓冲液p H、料液比、提取时间对PPO比活力影响显著。缓冲液p H与料液比交互作用极其显著,回归模型拟合性良好(R2=99.31%);影响显著顺序为:静置时间>缓冲液p H值>料液比;最优化工艺参数为:缓冲液p H7.5,料液比为1∶120,提取时间为14.5 min。在此条件下,酶比活力为137.389 U/mg。In this study, the method of acetone powder was optimized to extract the polyphenoloxidase ( PPO ) from honeysuckle flowers ( Lonicerajaponica ) by response surface methodology. The results showed that three operating parameters had great effects on the PPO specific activity, and their effects were ranked as follows : extraction time 〉 buffer pH 〉 material-to-liquid ratio. The established regression model was highly fitting ( R2 = 99. 31% ) , and the interaction effect between buffer pH and material-to-liquid ratio was extremely significant. The optimal extraction condition was pH 7. 49, material-to-liquid ratio 1 : 120 and extraction time 14. 5 min, under which the PPO specific activity was 137. 389 U/mg.
