摘 要:目的:通过观察香砂六君子丸对脾虚痰浊证大鼠小肠c AMP/PKA信号通路的影响,探讨香砂六君子丸治疗脾虚痰浊证大鼠可能机制。方法:24只SD大鼠,随机分为空白对照组、脾虚痰浊组、香砂六君子丸治疗组(以下简称治疗组),每组8只。采用高脂饲料喂养大鼠复制脾虚痰浊证动物模型,14 d后,治疗组采用香砂六君子丸灌胃治疗,空白对照组和脾虚痰浊模型组予等量生理盐水灌胃。灌胃60 d后采用ELISA法检测血浆中c AMP浓度,RT-PCR和Western blot检测大鼠小肠GP、PHK和PKA mRNA和蛋白表达水平。结果:与空白对照组相比,脾虚痰浊组小肠c AMP活性显著降低,与脾虚痰浊组相比,治疗组小肠c AMP活性显著升高;与空白对照组相比,脾虚痰浊组GP、PHK、PKA mRNA和蛋白含量显著下降,与脾虚痰浊组相比,治疗组GP、PHK、PKA mRNA和蛋白含量显著上升。结论:香砂六君子丸可能通过调控c AMP/PKA信号通路影响脾虚痰浊证大鼠。Objective:To observe the effect of Xiangsha Liujunzi Pill on small intestinal cAMP/PKA signaling pathway of rats with spleen deficiency and phlegm turbid and to explore the possible mechanism of Xiangsha Liujunzi Pill. Methods:24 SD rats were randomly divided into control group, spleen deficiency and phlegm turbid group and Xiangsha Liujunzi pill treatment group (hereinafter referred to as the treatment group) ,8 in each. The rats were fed with high fat diet to copy the spleen deficiency and phlegm turbid model. 14 d later,the treatment group was given Xiangsha Liujunzi Pill orally and control group and model group were given the same amount of normal saline by gavage. 60 days later,the ELISA assay was used for determining the plasma con- centrations of cAMP, RT - PCR and Western blot analysis for rat small intestine GP, PHK and PKA mRNA and protein expression levels. Results : Compared with the control group, spleen deficiency and phlegm turbid group intestinal cAMP activity was signifi- cantly decreased. Compared with the spleen deficiency and phlegm turbid group, the activity of intestinal cAMP in the treatment significantly increased. Compared with the control group,spleen deficiency and phlegm turbid group's GP,PHK,PKA mRNA and protein were significantly decreased. Compared with the spleen deficiency and phlegm turbid group, GP, PHK, PKA mRNA and protein levels of the treatment group increased significantly. Conclusion:Xiangsha Liujunzi Pill has effect on rats with spleen defi- ciency and phlegm turbid possibly through the regulation of cAMP/PKA signaling pathway.
