益气化瘀补肾方对压迫和减压后大鼠背根节神经元细胞凋亡的影响

摘　　要：目的:观察压迫和减压后大鼠背根节(DRG)组织中细胞凋亡及凋亡相关蛋白Bcl-2的表达变化,进一步探讨益气化瘀补肾方对上述指标的影响。方法:选用3月龄SPF级SD雄性大鼠50只,随机分为5组。分别应用TUNEL法检测DRG神经元凋亡程度,并应用免疫组织化学法检测凋亡相关蛋白Bcl-2的表达变化。结果:同假手术组相比,压迫组和减压组DRG神经元细胞凋亡个数显著增高,均具有统计学差异,P<0.01;减压后,凋亡细胞个数减少,与压迫组相比有显著性差异,P<0.05。压迫损伤后Bcl-2在DRG神经元表达升高,但与假手术组相比无显著性差异,P<0.05;在减压组Bcl-2表达进一步升高,与假手术组和压迫组相比,均有显著性差异,P<0.01或P<0.05。益气化瘀补肾方可显著降低DRG神经元凋亡的个数,促进Bcl-2表达,而在行减压手术后其作用更为显著,明显优于减压组,P<0.01。结论:益气化瘀补肾方可显著增加Bcl-2的表达,减少背根神经元的凋亡,其作用在减压手术后,更为显著。Objective：To observe the effect of Yiqi Huayu Bushen Decoction on apoptosis and apoptosis -related protein Bcl ＇ 2 of dorsal root ganglion neurons after compression and decompression model on rats. Methods ： Fifty 3 - week old healthy male SD rats were chosen and randomly divided into 5 groups. The apoptosis and apoptosis - related protein were measured by TUNEL and Immu- nohistochemistry. Results：Compared with the sham operation group,there was a significant increase in apoptosis of DRG in compres- sion group and decompression group （P 〈 0.01） and there was a more decrease in apoptosis of DRG in decompression group when compared with the compression group （P 〈 O. 05 ）. Compared with the sham operation group, there was no significant increase in the expression of BC1 -2 in the compression group （P 〈 0.05）, but it had an significant increase in decompression group compared with the sham operation group and compression group （P 〈 0.01 ,P 〈 0.05 ）. Yiqi Huayu Bushen Decoctioncould decrease the apoptosis and increase the expression of Bcl -2 of DRG and it could be more significant after removing the compression （P 〈 0.01 ）. Conclusion ： Yiqi Huayu Bushen Decoction could decrease the apoptosis and increase the expression of Bcl - 2 of DRG and it could be more significant after remove the compression.
