摘  要:目的分析比较5种WRI1转基因稻米粗脂肪表达效果。方法将椰子中的调节脂肪酸合成基因—WRI1基因导入粳稻品种中花11中,获得再生植株ZZWR1、ZZWR2、ZZWR3、ZZWR4和ZZWR5,用索氏提取法提取转基因稻米及其对照稻米中的脂肪,再将粗脂肪进行脂肪酸甲酯化,并用气相色谱法-质谱联用法(GC-MS)对其进行定性定量测定。结果与对照组的粗脂肪含量2.59%相比,ZZWR1、ZZWR2、ZZWR4的粗脂肪含量范围为2.62%~2.77%,其中ZZWR4的粗脂肪含量最高(2.77%),RSD=0.96%,与对照组相比差异有统计学意义(P〈0.01)。GC-MS测定棕榈酸甲酯、硬脂酸甲酯和油酸甲酯含量,其中油酸甲酯的含量最高。结论椰子中的WRI1基因的导入可提高稻米脂肪含量,但不同WRI1基因的插入位点可能会影响原有脂肪酸的表达,ZZWR4植株插入基因位点比较得当,具有良好的基因表达效果。Objective Analysis and comparison of crude fat expression effect of five kinds of WRH transgenic rice. Methods The coconut WRI1 gene (regulation genesof fatty acid synthesis ) was introduced into japonica rice zhonghuall, and 5 transgenie plants, ZZWR1, ZZWR2, ZZWR3, ZZWR4 and ZZWR5 were obtained. The crude fat of the transgenic rice and control rice was extracted by Soxhlet extraction method and determined qualitatively and quantitatively using GC-MS method. Results Compared with the control rice which contained 2.59% crude fat, the contents of ZZWR1, ZZWR2, ZZWR4 were from 2. 62% to 2.77% , which was significantly higher than the control rice. ZZWR4 was the highest with RSD of 0.96%. Palmitic acid methyl ester, methyl stearate and methyl oleate were quantified by GC-MS , and methyl oleate was the highest. Conclusion Coconut WRll gene introduced into rice could improve rice fat content. The different insertion sites of WRH gene may influence the original fatty acid expression. ZZWR4 insertion site had better expression.
