摘 要:目的:明确脂肪来源干细胞(adipose-derived stem cells,ADSCs)在辐射诱发的C57小鼠胸腺瘤的形成过程中是否通过增强DNA损伤修复功能进而影响辐射诱导胸腺瘤的形成。方法:清洁级C57BL/6小鼠32只,随机分为4组:对照组8只,仅以假照射;照射组8只,采用深部X射线治疗机照射;照射+ADSCs组8只,收集转染EGFP表达质粒48h后的ADSCs,将ADSCs的浓度调整为2.0×106个/mL,取0.5mL经尾静脉注射入末次照射后第1天的胸腺瘤模型小鼠;假照射+ADSCs组8只,ADSCs的注射方法同照射+ADSCs组。末次照射第7天后分别处死4组小鼠,各组小鼠胸腺组织中的细胞周期蛋白D1(cyclin-D1)、p53、c-myc、毛细血管共济失调突变期(ATM)和γ-H2AX蛋白水平采用Western blot进行检测。结果:照射组小鼠胸腺组织中ATM、γ-H2AX、cyclin-D1和c-myc水平高于照射+ADSCs组小鼠胸腺组织,而p53水平低于照射+ADSCs组小鼠胸腺组织,且差异有统计学意义(P<0.05)。通过流式细胞术检测,照射组小鼠胸腺G1期细胞的百分比相比于对照组明显降低(P<0.05),而照射+ADSCs组小鼠胸腺G1细胞的百分比较照射组增多。结论:ADSCs可能通过减轻DNA损伤程度,减少p53和c-myc蛋白的表达,使细胞G1期增加,并阻滞在G1期,修复胸腺细胞的DNA,抑制肿瘤的形成。Objective:To investigate the effects of compoundα-keto acid tablets on nutritional status,calcium-phosphorus metabolism and inflammatory factors in patients with maintenance hemodialysis.Methods:84patients with maintenance hemodialysis were enrolled and randomly divided into observation group and control group with 42 cases in each group.The control group were treated with maintenance hemodialysis,the observation group were treated with compoundα-keto acid tablets based on treatments of the control group.The serum total protein(TP),serum albumin(ALB),hemoglobin(HB),transferrin(TRF),calcium(Ca),phosphorus(P),calcium phosphorus product(Ca×P),serum intact parathyroid hormone(iPTH),interleukin 6(IL-6),tumor necrosis factor alpha(TNF-alpha)and high sensitive C reactive protein(hs-CRP)of two groups were tested before and after treatment.Results:After treatment,the TP,ALB,HB,TRF of observation group were significantly higher than those in the control group(P0.05),and blood P,blood Ca×P,serum iPTH were significantly lower than the control group(P0.05),serum IL-6,TNF-alpha and hs-CRP levels were significantly lower than those of the control group(P0.05).Conclusion:Compoundα-keto acid tablets can effectively improve the nutritional status of hemodialysis patients,maintain the metabolism of calcium and phosphorus balance,and can reduce the inflammatory reaction,ant it is worth the clinical promotion and application.
