摘 要:目的检测同源异型盒基因D8(HOXD8)在人卵巢癌非顺铂耐药株SKOV3和顺铂耐药细胞株SKOV3-DDP细胞株中的表达,探讨卵巢癌顺铂耐药的机制。方法应用细胞活性测定实验(XTT实验)检测SKOV3、SKOV3-DDP细胞株经顺铂处理后的半数抑制浓度,荧光定量PCR、ELISA检测HOXD8在这两种细胞株中的表达。结果随着顺铂浓度的增加,细胞抑制率增高,显示出明显的剂量依赖关系;SKOV3-DDP细胞株经顺铂作用后,细胞抑制率明显低于SKOV3细胞株。荧光定量PCR及ELISA试验结果显示,与SKOV3细胞株比较,SKOV3-DDP细胞株中HOXD8mRNA及蛋白的相对表达量均明显升高,差异有统计学意义。结论 HOXD8表达的升高与卵巢癌细胞顺铂耐药存在明确的相关性。Objective To detect the expression of homeobox genes D8 (HOXD8) in human ovarian cancer SKOV3 and SKOV3-DDP cell line. To investigate the mechanism of cisplatin-resistance in ovarian cancer. Methods XTT was used to detect the half inhibitory concentration after treatment of cisplatin between SKOV3 and SKOV3-DDP cell line. Fluorescence quantita- tive PCR and ELISA were used to detect the expression of HOXD8 in human ovarian cancer SKOV3 and SKOV3-DDP cell line. Results As the concentration of cisplatin increased, the inhibition rate increased. It showed obvious dose dependence. The in- hibition rate was significantly lower in SKOV3-DDP than SKOV3 cell line after treatment of cisplatin. PCR and ELISA test re- sults showed that HOXD8 mRNA and protein expression levels were obviously increased, compared with SKOV3 cell lines, which was statistically significant. Conclusion There is a clear correlation between HOXD8 expression and cisplatin resistance in ovarian cancer cells.
