摘  要:目的:通过检测口腔癌及癌前病变中错配修复基因MLH-1与抑癌基因PTEN的蛋白表达并分析二者之间的相关性及意义.方法:用免疫组织化学SP法检测48例口腔鳞状细胞癌、上皮单纯增生11例及上皮异常增生10例,而正常口腔黏膜10例中错配修复基因MLH-1与抑癌基因PTEN的蛋白表达水平,并对结果进行统计分析.结果:口腔鳞状细胞癌中MLH-1的阳性表达率为47.9%(23/48),明显低于其他各组;PTEN蛋白的阳性表达率为47.9%(23/48),明显低于其他各组,与正常黏膜、上皮单纯增生、上皮异常增生对比均有显著性差异(P<0.05).经直线相关分析和Spearman等级相关分析,MLH-1与PTEN之间存在正相关的关系(r=0.548,P=0.018).结论:MLH-1与PTEN在口腔鳞状细胞癌发生过程中均起作用,MLH-1与PTEN可能存在正相关的关系.Aim: The purpose of this study was to investigate the expression of MLH-1 and PTEN proteins in oral squamous cell carcinomas( OSCC) and precancerous lesions,discuss the significance,and analyze the correlation between them. Methods: Immunohistochemical SP method was used to detect the expression of MLH-1 and PTEN in 48 cases of oral squamous cell carcinoma,11 cases of simple hyperplasia,10 cases of DYSplasia,and 10 cases of normal oral mucosa. Results: The positive rate of MLH-1 in48 cases of OSCC( 47. 9%) was significantly lower than that in non-malignant groups,which included normal oral mucosa,DYS and simple hyperplasia( P < 0. 05),while the positive rate of PTEN in 48 ca-ses of OSCC( 47. 9%) was significantly lower than that in non-malignant groups( P < 0. 05). A positive correlation was found between the expressions of MLH-1 and PTEN( r = 0. 548,P = 0. 018) in linear correlation analysis and Spearman correlation test. Conclusion: Our data suggested that MLH-1 and PTEN played important roles in the pathogenesis of OSCCs,and there may be positive correlation between them.
