摘 要:目的探讨苯肾上腺素预处理对放射损伤早期大鼠颌下腺p-JNK表达的影响。方法取Wistar大鼠27只,随机分为空白对照组、单纯照射组和苯肾上腺素预处理组,预处理组大鼠照射前半小时腹腔注射苯肾上腺素5mg/kg。应用直线加速器给予大鼠颌下腺区域20Gy X线照射,照射后7天取大鼠颌下腺组织,应用免疫组织化学方法检测各组大鼠颌下腺p-JNK(phospho)的表达及定位;应用免疫印迹法(Western blot)检测各组大鼠颌下腺JNK(c-Jun N-terminal kinase JNK)及p-JNK的表达。结果免疫组织化学结果显示:空白对照组颌下腺腺泡及导管细胞的胞浆和胞核中仅有极少量p-JNK的表达,单纯照射组p-JNK在腺泡及导管细胞的胞浆和胞核中的表达明显增加,而苯肾上腺素预处理组p-JNK在腺泡及导管细胞的胞浆和胞核的表达较单纯照射组明显减少。Western blot结果显示:单纯照射组p-JNK蛋白的表达较空白对照组增加了44.00%(P<0.05),苯肾上腺素预处理组较单纯照射组下降了64.16%(P<0.01),各组JNK蛋白的表达未见明显差异(P<0.05)。结论 X线照射损伤早期大鼠颌下腺p-JNK的表达增加,苯肾上腺素能够下调损伤后大鼠颌下腺p-JNK的表达,提示苯肾上腺素促放射损伤后涎腺腺体修复的机制可能与活化1-肾上腺素受体下调p-JNK的表达有关。Objective To investigate the effect of phenylephrine pretreatment on the expression of p-JNK in irradiated submandibular gland in rat during the early stage. Methods Twenty-seven Wistar rats were divided into three groups by chance and respectively named: the control group, the irradiation-only group, and the phenylephrine-pretreatedgroup, phenylephrine(5mg/kg) was injected intraperitoneally 30 min prior to irradiation. 20 Gy X-ray irradiation was delivered to the submandibular region by applying a medical linear accelerator, and the submandibular glands were collected on day 7 after irradiation. The distribution and expression of p-JNK was then detected by immunohistochemical assay, and Western blot was applied to detect the expression of p-JNK and JNK. Results The expresssion of p-JNK was mainly founded in the cytoplasm and nucleus of both serous acinar and ductal cells in the control group, compared with the control group, it was significantly increased in the irradiation-only group, and in the phenylephrine- pretreated group the distribution was markedly decreased. Also, results from Western blot demonstrated that there was no obvious difference of the expression of JNK between the three groups, the expression of p-JNK was upregulated 44.00%(P〈0.05) in the irradiation-only group compared with the control group, but was down- regulated64.16.%(P〈0.05) in the phenylephrine-pretreated group compared with the irradiation-only group. Conclusion The expression of p-JNK in the rat submandibular gland could be increased after irradiation during the early stage and decreased by phenylephrine pretreatment. The results indicated that the mechanism of phenylephrine pretreatment improving salivary gland recovery after irradiation damage might be involving in the decreased expression of p-JNK by activation of α1-adrenergic receptor, which might provide a new strategy in preventing and renewing salivary gland irradiated damage.
