紫草素干预下金黄地鼠血清对口腔癌KB细胞增殖活性的影响

摘　　要：目的研究紫草素干预下金黄地鼠血清对口腔癌KB细胞增殖活性的影响。方法Ⅰ组:以含20%灭活普通金黄地鼠血清培养液培养口腔癌KB细胞;Ⅱ组:以含20%灭活紫草素干预金黄地鼠血清培养液培养口腔癌KB细胞;用MTT比色法、流式细胞术测定两组细胞生长曲线、细胞DNA合成以及细胞周期分布,并计算各个时间点抑制率、细胞S期分数和增殖指数。结果比较两组不同时间点OD值,两组间有显著差异性(P<0.05)。两组不同时间点抑制率结果显示差异有统计学意义(P<0.05)。Ⅱ组细胞G0/G1期细胞所占的比例高于Ⅰ组,而S期及G2/M期细胞所占的比例明显低于Ⅰ组;Ⅱ组S期分数(t=3.548,P<0.05)及细胞增殖指数(t=2.731,P<0.05)显著低于Ⅰ组,差异有统计学意义。结论长期紫草素干预条件下金黄地鼠血清能抑制口腔癌KB细胞的生长以及DNA合成。紫草素对口腔鳞癌KB细胞的增殖具有明显的抑制作用。Objective To study the effects of Golden Hamster serum on the proliferation activity of oral cancer KB cells under the interference of Shikonin. Methods Group Ⅰ ：Incubate the oral cancer KB cells in a kind of incubation solution containing 20deactivated general Golden Hamster serum; Group Ⅱ ： Incubate the oral cancer KB cells in a kind of incubation solution containing 20deactivated general Golden Hamster serum with Shikonin interference; The cell growth curve, the cellular DNA synthesis and the cell-cycle distribution were determined through the MTT colorimetric method and the flow cytometry for the two groups, and the inhibition rate, the cell S-phase fraction and the proliferation index were calculated for each time point. Results The OD value was compared at each time point between the two groups and there were significant differences between the two groups（P〈0.05）. There were statistically significant differences in the inhibition rate results at different time points between the two groups（P〈0.05）. The proportion of G0/G1-phase cells was higher in Group Ⅱ that that in Group Ⅰ, and the proportions of S-phase and G2/Mphase cells were significantly lower than those in Group I. The S-phase fraction（t =3.548, P〈0.05） and the cell proliferation index（t=2.731, P〈0.05） in Group Ⅱ were significantly lower than those in Group I, and the differences were statistically significant. Conclusion With long term Shikonin interference the Golden Hamster serum can inhibit the growth of and DNA synthesis in oral cancer KB cells. Shikonin has significant inhibition on the proliferation of oral squamous cell carcinoma KB cells.
