摘 要:目的:评价可注射纳米羟基磷灰石/半水硫酸钙(nano-hydroxyapatite/collagen/calcium sulphate hem ihydrate,nHAC/CSH)植骨材料对犬外周血基质细胞(dog peripheralblood strom alcells,dPBSCs)骨向分化的影响。方法:体外培养犬外周血基质细胞,分别用成骨诱导液和nHAC/CSH浸提后的成骨诱导液培养dPBSCs,检测ALP活性,利用RT-PCR检测成骨基因Runx2、Col I、OPN的表达;采用Western Blot检测Runx2、Col I、OPN蛋白表达的水平。结果:在各时间点nHAC/CSH浸提液培养的d PBSCs ALP活性(4d:11.801±0.614;7d:20.589±0.693)高于成骨诱导液组(4d:5.048±0.674;7d:7.690±1.852);7天时浸提液组OPN mRNA表达明显高于成骨诱导液组;浸提液组R unx2、OPN蛋白表达的灰度值明显高于成骨诱导液组。结论:nHAC/CSH在一定程度上提高d PBSCs向成骨细胞分化的能力。Objective: To assess effects of the injectable nano-hydroxyapatite/collagen/calcium sulphate hemihydrate(nHAC/CSH) on osteogenic differentiation of peripheral blood stromal cells(dPBSCs). Methods: Peripheral blood stromal cells were isolated from peripheral blood of healthy adult dogs and cultured in the extract medium(EX) and osteogenic medium(C). The ALP activity was measured. The level of Runx2、 Col I、 OPN mRNA expression was measured using RT-PCR. The expression of Runx2、Col I、OPN protein was detected by Western blot analysis. Results: The cells in the extract medium showed a higher ALP activity than those of the d BMPCs in the osteogenic medium at every time point tested(P〈0.05). The expression of OPN mRNA in EX group was higher than that in the C group. The protein expressions of Runx2 and OPN in EX group were higher than those in the C group. Conclusion: n HAC/CSH could improve the ability of osteogenic differentiation of dPBSCs.
