摘　　要：目的本研究旨在研究JAK/STAT途径的负调控因子,细胞因子信号抑制因子1(SOCS-1)在宫颈癌中的作用。方法收集60份宫颈癌新鲜手术标本和20份非肿瘤标本,用real-time RT-PCR、Western blot和免疫组化实验分析宫颈癌不同病理阶段SOCS-1 mRNA和蛋白的表达、高危HPV感染的标本和无HPV感染的SOCS-1的表达,并与正常宫颈组织进行比较。结果与正常组织中SOCS-1比较,58%的肿瘤组织不表达或减少表达,免疫组化检测、realtime RT-PCR和Western blot的结果一致。与未感染HPV的标本比较,高危HPV感染的标本低表达或不表达SOCS-1。结论高危型HPV16、HPV18的感染可导致SOCS-1的转录失活,表明高危型HPV癌蛋白抑制SOCS-1的表达,意味着SOCS-1在宫颈癌发展中起重要作用。Objective This study was designed to study the role of negative regulatory factors with JAK / STAT pathway and the suppressor of cytokine signalling- 1（ SOCS- 1） in cervical cancer. Methods To collect 60 cervical cancer fresh surgical specimens and 20 cases of non cancer specimens. Using real- time reverse transcriptase- polymerase chain reaction（ real- time RT- PCR）,Western blot and immunohistochemistry experiment,to analyze the expression of SOCS- 1 mRNA and protein in different pathological stages of cervical cancer,the expression of HPV and SOCS- 1 infection in high risk HPV infection,and to compare them with that in normal cervical tissue. Results Compared with expression of SOCS- 1 in normal tissues,58% of the tumor tissues expressed either undetectable or reduced expression. The results of immunohistochemistry assay,real- time RT- PCR and Western blot were the same. The SOCS- 1 levels expressed undetetable or reduced expression in high- risk-HPVs were compared with one in no HPV infection samples. Conclusion We demonstrate that the transcriptional inactivation of SOCS- 1 gene due to HPV16 and HPV18 infection,suggesting that SOCS- 1 may play an important role in cervical carcinoma.
