摘  要:目的建立超高效液相色谱-串联质谱法(UPLC-MS/MS)测定火腿肠中红2G工业染料含量的方法。方法样品中的红2G染料经过乙腈-水(7∶3,V/V)溶液浸泡、超声提取后,低温高速离心、旋转蒸发浓缩,再用ACQUITY UPLC BEH C18(100 mm×2.1 mm,1.7μm)色谱柱,以乙腈-乙酸铵(20 mmol/L)溶液为流动相进行梯度洗脱,采用电喷雾负离子(ESI-)模式进行测定。结果红2G的浓度为5 ng/ml^100 ng/ml时,具有良好的线性关系,回归方程为y=37.7x+18.5,相关系数(r)=0.999 2,该方法的检出限为0.8μg/kg,定量限为2.6μg/kg。在10.0μg/kg、50.0μg/kg和100.0μg/kg 3种添加水平下,红2G工业染料的平均加标回收率为87.9%~90.0%,相对标准偏差为1.1%~2.5%。结论该方法操作快捷、灵敏度高、结果准确可靠,适用于火腿肠中红2G工业染料的快速测定和确证。Objective A method was established for the determination of Red 2G industrial dye content in sausage using ultra-high performance liquid chromatography- tandem mass spectrometry( UPLC- MS / MS). Methods Red 2G dye in the samples was soaked and ultrasonically extracted by acetonitrile- water( 7∶3,V / V),then the extract was purified by super centrifuge in low temperature,concentrated by rotary evaporation,separated by ACQUITY UPLC BEH C18column( 100 mm × 2. 1 mm,1. 7 μm) with acetonitrile and ammonium acetate( 20 mmol / L) as mobile phase for gradient elution and to detect by the negative ion( ESI-) model. Results The method showed a good linearity in the range of 5 ng/ml- 100 ng/ml with the correlation coefficients( r) greater than 0. 999 2,and the regression equation is y = 37. 7x + 18. 5. The detection limit of this method was0. 8 μg / kg,and the limit of quantification was 2. 6 μg / kg. The average additive recoveries rate of Red 2G industrial dye at three levels( 10. 0 μg / kg、50. 0 μg / kg and 100. 0 μg / kg) were 87. 9%- 90. 0% with relative standard deviations were1. 1%- 2. 5%. Conclusion This method exhibits easy operation,high sensitivity and accuracy. It is suitable for the rapid determination and confirmation of Red 2G industrial dye in sausage.
