摘　　要：目的探讨PDCD4对肺癌细胞增殖、凋亡的影响及分子机制。方法利用QPCR检测非小细胞肺癌组织和远处肺组织中PDCD4表达量。利用化学合成的PDCD4 siRNA转染A549及PC9肺癌细胞株,通过MTT及流式细胞仪Annix V-FITC法检测细胞生长与凋亡,观察干扰PDCD4后细胞生长及凋亡的变化;利用Western blot检测细胞周期和细胞凋亡相关蛋白。结果 QPCR结果显示,PDCD4在非小细胞肺癌组织中低表达(P=0.008)。MTT法显示,与对照组相比,干扰PDCD4组细胞生长速度增加(P<0.01)。流式细胞仪Annix V-FITC法检测细胞凋亡显示,干扰PDCD4组细胞凋亡的数量减少(P<0.05)。Western blot结果显示,与对照组相比,干扰PDCD4的A549和PC9细胞中,CCND1和Bcl-xl表达上调,P15和cleaved-caspase 3表达下调。结论 PDCD4在非小细胞肺癌组织中低表达,且PDCD4可能通过下调CCND1和Bcl-xl的表达,上调P15和cleaved-caspase 3的表达抑制肺癌细胞株A549和PC9增殖并诱导其凋亡。Objective To explore the effect and molecular mechanism of PDCD4 on human lung cancer cell proliferation and apoptosis. Methods Expression of PDCD4 mRNA in NSCLC tissues and distal paracarcinoma tissues was detected by QPCR. PDCD4 siRNAs were synthesized and transfected into lung cancer A549 and PC9 cells. The abilities of cell growth and apoptosis were detected by MTT and Annexin V-FITC by FACS. Western blot was used to examine the expression of cell cycle- and cell apoptosisassociated proteins. Results Compared to the distal paracarcinoma tissues,PDCD4 was down-regulated in NSCLC tissues（ P = 0. 008）. Compared with the control group,PDCD4-knocked down elevated cell growth（ P 0. 001） and inhibited cell apoptosis（ P 0. 05） in A549 and PC9 cells. Meantime,PDCD4-knocked down obviously increased the expression of CCND1 and Bcl-xl,and decreased the expression of P15 and cleaved-caspase 3. Conclusion PDCD4 is decreased in NSCLC tissues compared with distal paracarcinoma tissues. PDCD4 may suppress cell proliferation and induce cell apoptosis in A549 and PC9 cells,which is related to down-regulation of CCND1 and Bcl-xl expression and enhancement of P15 and cleaved-caspase 3expression.
