摘  要:目的:通过脂质转染mi R-181b-5p模拟物及抑制物到卡波西肉瘤细胞系SLK,首次研究mi R-181b-5p对卡波西肉瘤细胞SLK细胞增殖、凋亡生物学功能的影响,为进一步研究mi R-181b-5p在卡波西肉瘤发病机制中的作用奠定理论基础。方法:应用脂质体对人卡波西肉瘤细胞株SLK进行转染,对转染mi R-181b-5p模拟物及抑制物后的卡波西肉瘤细胞株行MTT检测观察细胞增殖曲线;应用流式细胞仪行检测转染48小时后各组细胞凋亡率比较。结果:与阴性对照组比较,mi R-181b-5p模拟物组细胞的增殖能力明显上升,(P<0.01)。mi R-181b-5p抑制物组细胞的增殖能力明显下降,(P<0.01)。mi R-181b-5p抑制物阴性对照组细胞的增殖能力无明显变化。流式细胞仪测定SLK细胞转染mi R-181b-5p模拟物后凋亡率(5.5±0.6)%,与阴性对照组凋亡率(7.6±0.4)%相比,差异有统计学意义,明显下降(P<0.05);mi R-181b-5p抑制物组凋亡率(14.8±1.0)%明显增高,有统计学差异(P<0.05)。结论:通过转染mi R-181b-5p模拟物/抑制物,发现其对SLK细胞的增殖、凋亡造成一定影响,进一步开展mi R-181b-5p生物学特性和其相关靶基因验证的相关研究有十分重要的意义。Objective: miR-181b-5p mimics and inhibitors were transfected into Kaposi's sarcoma cell line SLK through liposomal transfection. The effect of miR-181b-5p on SLK cell proliferation and apoptosis of biological activities were studied to provide theoretical basis for further investigation on the role of miR-181b-5p in the pathogenesis of Kaposi's sarcoma. Methods: Human Kaposi's sarcoma cell line SLK was transfected by liposomes. The proliferation of SLK cell was assessed by MTT assay; apoptosis rates of each group were determined by flow cytometry at 48 hours after transfection. Results: Compared to the negative control group, proliferative capacity of miR- 181 b-5p mimics has increased significantly after transfection (P 〈 0.01), proliferative capacity of miR- 181 b-5p inhibitor has de- creased significantly after transfection (P 〈 0.01). Flow cytometry showed that, apoptosis rate of the SLK cells has decreased significantly after transfection with miR-181b-5p mimics (5.5± 0.6) % compared to that of corresponding negative control (7.6± 0.4) % (P〈0.01). Apoptosis rate of the SLK ceils has increasedsignificantly after transfection with miR-181b-5p inhibitor (14.8± 1.0)compared to that of corresponding negative control(7.8 ± 0.5) % (P 〈 0.01). Conclusions: Transfection with miR- 181 b-5p mimics/inhibitors significantly af- fected SLK cell proliferation and apoptosis. Further researches on biological characteristics of miR-181 b-5p and validation of the associ- ated target genes are therefore of great significance.
