摘  要:目的:探讨Src激酶特异性抑制剂PP2对人胆管癌QBC939细胞侵袭能力的影响和机制。方法:通过Western Blotting技术检测PP2对人胆管癌QBC939细胞中Src激酶活化的影响;用Transwell小室法观察PP2对QBC939细胞的影响;用RT-PCR和Western Blotting技术检测PP2对QBC939细胞侵袭能力相关分子的作用。结果:实验组p-Src蛋白表达水平明显低于对照组,差异具有统计学意义(P<0.05);实验组QBC939细胞体外侵袭能力较对照组显著降低,差异具有统计学意义(P<0.05);与对照组相比,实验组E-cadherin表达显著增强,CD44表达明显减弱,差异具有统计学意义(P<0.05)。结论:PP2通过抑制Src激酶活化,增强E-cadherin表达、减弱CD44表达,抑制人胆管癌QBC939细胞侵袭能力。Objective: To detect the effect of Src kinase inhibitor PP2 on the invasion of human cholangiocarcinoma QBC939 cells. Methods: Western blotting was used to detect the effect of Src kinase inhibitor PP2 on Sre activation. Transwell chamber were performed to detect the effect of Src kinase inhibitor PP2 on the invasion of human cholangiocareinoma QBC939 cells. The effect of Sre kinase inhibitor PP2 on the invasion-related molecules of human cholangiocareinoma QBC939 cells by RT-PCR and western blotting. Results: The expression ofp-Sre protein in PP2 group was significantly lower than control group. The invasion of QBC939 cell in PP2 group was obviously decreased than control group. And compared with control group, E-cadherin of PP2 group expression was observably enhanced, CD44 expression was dramaticUy decreased. Conclusion: PP2 can inhibit Sre kinase activation, increase the expression of E-cadherin, reduce the expression of CD44, then inhibit the invasion of QBC939 human cholangiocarcinoma cells.
