摘  要:目的:探讨使用荧光原位杂交技术检测乳腺原发癌和淋巴结转移癌HER-2基因的表达及意义。方法:选取我院2013年6月至2014年6月收治的乳腺原发癌患者133例作为研究对象,对其临床资料进行回顾性分析。应用荧光原位杂交技术(FISH)与免疫组化技术(IHC)对所有患者手术切除标本的HER-2基因表达予以检测。结果:FISH阳性率为42.1%,IHC为58.6%;IHC阴性时与FISH符合率为87.2%,IHC(+)与FISH符合率为34.7%,IHC(++)与FISH符合率为83.8%,IHC(+++)与FISH符合率为100.0%。淋巴结转移阳性率为67.5%,未转移阳性率为33.3%,组间比较,差异具有统计学意义(P<0.05)。结论:IHC对HER-2基因予以检测时若结果为阴性或者强阳性则与FISH符合率较高,同时HER-2基因表达与腋淋巴结转移之间存在正相关关系。Objective To investigate the expression and significance of using fluorescence in situ hybridization detection of primary breast cancer and lymph node metastasis of HER-2 gene. Methods in our hospital from 2013 June to 2014 June were analyzed in 133 cases of primary breast cancer patients as the research object, and the clinical data were retrospectively analyzed. Application of fluorescence in situ hybridization(FISH) and immunohistochemical technique(IHC) to detect the expression of HER-2 gene in all patients of resected specimens. Results The positive rate of FISH was 42.1 %, IHC was 58.6 %; IHC negative and FISH coincidence rate was 87.2 %, IHC(+) and FISH coincidence rate was 34.7 %, IHC(+ +) and FISH coincidence rate was 83.8 %, IHC(+ + +) and FISH coincidence rate was 100 %. Lymph node metastasis positive rate was 67.5 %, not to transfer the positive rate was 33.3 %, compared difference was statistically significant(P〈0.05). Conclusion IHC to be detected HER-2 gene if the result is negative or positive and strong FISH coincidence rate is higher, at the same time, the expression of HER-2 gene and there is a positive correlation between axillary lymph node metastasis.
