摘  要:目的:探讨人参多糖注射液对人乳腺癌细胞株MCF-7侵袭迁移能力的影响。方法以不同作用浓度(12.5μL/mL、25μL/mL、50μL/mL、100μL/mL)人参多糖注射液处理MCF-7细胞株,以PBS作为空白对照,分别采用Transwell小室侵袭实验、划痕实验及Western blot检测细胞侵袭能力、迁移能力及基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)表达。结果人参多糖注射液对MCF-7细胞体外侵袭和迁移能力具有显著的抑制作用,并呈现剂量及时间依赖性。人参多糖注射液处理后,MCF-7细胞MMP-2蛋白表达及MMP-9蛋白表达比值显著降低,并呈现剂量及时间依赖性。结论人参多糖注射液可显著抑制人乳腺癌细胞MCF-7侵袭能力和迁移能力,下调MMP-2及MMP-9蛋白表达是其可能的作用机制。Objective To evaluate the effect and mechanism of ginseng polysaccharides injective solu-tion on invasive ability of human breast cancer MCF-7 cells.Methods Human breast cancer MCF-7 cells were treated with different concentrations of ginseng polysaccharides injections(12.5μL/mL,25μL/mL,50μL/mL or 100μL/mL),and phosphate buffer saline was served as control.The invasive ability,transfer ability and protein expression of matrix metalloproteinases-2 ( MMP-2 ) and matrix metalloproteinases-9 ( MMP-9 ) were determined via transwell matrigel invasive test,scratch test and Western Blot,respectively.Results Ginseng pol-ysaccharide injection on invasive ability and transfer ability in vitro of MCF-7 cell had significant role in inhibi-ting effect,and showed dose and time dependent.After ginseng polysaccharides injection treatment,the MMP-2 and MMP-9 protein expression significantly reduced,and showed dose and time dependent manner.Conclusion Ginseng polysaccharides injection is able to inhibit the invasion and transfer in MCF -7 cells.Moreover,theregulation of MMP -2 and MMP -9 expression may be involved in this effect.
