摘  要:目的探讨NDRG2(N-Myc downstream-regulated gene 2)调控肝星形细胞活化的方式及机制。方法通过细胞免疫荧光和Western blot的方法检测肝星形细胞中NDRG2的定位及表达;利用腺病毒载体上调肝星形细胞中NDRG2的表达,检测高表达NDRG2对肝星形细胞活化的影响;Western blot检测TGF-β1/Smad通路,阐明NDRG2调控肝星形细胞活化的机制。结果 NDRG2在肝星形细胞的胞质中表达,其表达水平在肝星形细胞活化的过程中逐渐下降;利用腺病毒载体上调肝星形细胞中NDRG2的表达能够抑制肝星形细胞的活化;肝星形细胞活化后,Smad3的蛋白表达增加,而NDRG2能够使Smad3的表达降低。结论 NDRG2通过影响TGF-β1/Smad通路调控肝星形细胞的活化。Objective To investigate the distribution and expression pattern of N-Myc downstream-regulated gene 2 (NDRG2) in hepatic stellate cells(HSCs),and to explore its role and regulation mechanism in the HSCs activation process.Methods Immunofluorescence analysis and Western blot were used to detect the distribution and expression pattern of NDRG2 in HSCs.The effect of NDRG2 overexpression in HSCs on HSCs activation was investigated.TGF-β1/Smad pathway was detected by Western blot to explore the regulation mechanism of NDRG2 in the HSCs activation process.Results NDRG2 located in the cytoplasm of HSCs.NDRG2 protein levels were gradually reduced during HSCs activation.NDRG2 overexpression inhibited HSCs activation.Smad3 protein was increased after HSCs activation.The enhanced NDRG2 expression reduced Smad3 expression.Conclusion NDRG2 could regulate the HSCs activation by inhibiting TGF-β1/Smad signaling pathway.
