摘 要:目的建立一个合适的人乳腺癌裸鼠移植动物模型,拟研究移植瘤的病理特征。方法采用雌激素受体阳性的MCF-7人乳腺癌细胞株,接种于20只裸小鼠右下腹壁,观察肿块生长速度、瘤体表面皮肤受损情况,在第45天处死荷瘤裸鼠。结果接种后第10天在接种部位可见瘤结节,肿瘤移植成功率90.0%(18/20),第20-30天肿瘤生长较快,第45天处死荷瘤裸鼠,切除肿块,肿瘤平均直径和质量分别为(11.08±1.38)mm和(0.996±0.097)g;肿块表面皮肤完整。第26天开始小鼠出现死亡现象,至第45天时死亡8只,死亡率40.0%(8/20)。瘤体病理学检查为浸润性导管癌,雌激素受体ER阳性。结论本研究成功将乳腺癌MCF-7(ER+)细胞株植入裸鼠右下腹壁,肿瘤生长良好、肿瘤表面皮肤完整。乳腺癌的病理特征得以保留。这可能是开展乳腺癌研究的一个有效的模型。Objective To establish a proper model of human breast carcinoma in nude mice for studying their biological features.Methods MCF-7 cells(ER +) were implanted into the right lower abdominal wall in 20 nude mice.Tumor growth rate and the skin damage of tumor surface were observed,and the tumor-bearing mice were killed at day 45.The tumor masses were removed and pathologically observed under microscope.ER protein positive expressim was observed by immtmohistochemistry staining.Results The tumor tubercles were found at the incubated sites at day 10.The successful rate of implanted tumor was 90.0%(18/20).The tumor grew rapidly at days 20-30.At day 45,the tumor bearing nude mice were executed,and the average diameter and weight of the tumors were(11.08 ± 1.38) mm and (0.996 ± 0.097) g,respectively.The tumor surfaces were intact.The nude mice began to die from the 26th day,and 8 died until the day 45.The death rate was 40.0%(8/20).All of the specimens were infiltrating ductal carcinoma with estrogen receptor positive.Conclusion MCF-7 cells (ER +) are implanted into the right lower abodominal wall to successfully establish the human breast carcinoma in nude mice.The tumor grows well and the tumor surfaces are intact.Certain biological features of human breast carcinoma are reserved in the tumor.It may be an available model for further research of human breast carcinoma.
