摘  要:目的探究大蒜素对大鼠心肌缺血的保护作用及其相关机制。方法将50只准性SD大鼠随机平均分成假手术组(SO组),缺血/再灌注组(I/R组),大蒜素处理组高剂量组(HA组)、中剂量组(MA组)、低剂量组(LA组),每组10只。大蒜素处理组分别灌胃给予100、50、25 mg/(kg·d)大蒜素,给药20 d后构建大鼠心肌缺血/再灌注模型。检测各组大鼠血清及缺血心肌组织肌酸激酶同工酶(creatine kinase-MB,CK-MB),丙二醛(malondialdehyde,MDA),超氧化物歧化酶(superoxide dismutase,SOD)含量;Tunel法检测各组大鼠心肌组织细胞凋亡情况;免疫印迹法检测天冬氨酸蛋白水解酶-3(aspartic protease-3,caspase-3),磷酸化分裂原激活的蛋白激酶(phosphorylation of mitogen activated protein kinase,p-p38MAPK)及分裂原激活的蛋白激酶(mitogen-activated protein kinase,p38MAPK)的表达情况。结果 HA组与MA组的CK-MB、MDA水平均比I/R组显著偏低(P<0.05),HA组的SOD水平较I/R组显著偏高(P=0.015)。HA组,MA组的心肌损伤范围及凋亡指数较I/R组显著偏低(P<0.05),HA组caspase-3蛋白表达水平较I/R组显著偏低(P=0.023)。5组p38MAPK蛋白表达量无显著差异,但是HA组p-p38MAPK蛋白表达量较I/R组显著偏高(P=0.038)。结论大蒜素对大鼠心肌缺血具有明显的保护作用,且这种保护作用可能与心肌凋亡的抑制作用及p38蛋白的活化具有一定关系。Objective To investigate the protective effect of allicin on myocardial ischemia in rats and related mechanisms. Methods 50 SD rats were randomly divided into sham operation group( SO group),ischemia / reperfusion group( I / R group),allicin treatment groups: high dose allicin group( HA group),middle dose allicin group( MA group),low dose allicin group( LA group),10 rats in each group. 100,50,25 mg /( kg·d) allicin were administered into rats in allicin treatment groups for 20 days,respectively. All groups except SO group were constructed of myocardial ischemia /reperfusion( I / R) model. Serum and ischemic myocardial tissue were obtained to examine creatine kinase-MB( CK-MB),malondialdehyde( MDA),and superoxide dismutase( SOD) by the corresponding kit; cardiac tissue were used to detect damage extents and cell apoptosis by Tunel method;myocardial protein was extracted and the expression of aspartic protease-3( caspase-3), phosphorylation of mitogen activated protein kinase( p-p38MAPK) and mitogen-activated protein kinase( p38MAPK) were detected by Western blot. Results Levels of CK-MB and MDA in HA group and MA group were lower than I / R group( P〈0. 05),and level of SOD was higher( P = 0. 015). Myocardial injury extent and apoptosis index in HA group and MA group were smaller than that in I / R group( P〈0. 05). Caspase-3 protein expression level in HA group was lower than that in I / R group( P = 0. 023). No significant difference was detected among the five groups in p38 MAPK protein expression,but HA group p-p38 MAPK protein expression level was significantly higher than I / R group( P = 0. 038). Conclusion Allicin has a significant protective effect on myocardial ischemia,and may have some relationship with the inhibition of myocardial apoptosis and p38 MAPK activation.
