摘  要:核因子E2相关因子2(nuclear factor E2 related factor 2,Nrf2)是机体抗氧化应激的中枢调节者。在正常情况下,其在胞质中被降解,不发挥生理作用。当其在体内被有毒有害物质激活后转位进入细胞核能与抗氧化反应元件(antioxidant response element,ARE)结合形成Nrf2-ARE信号通路,从而使下游一系列具有机体保护性的Ⅱ相解毒酶和抗氧化酶基因及蛋白得以表达,主要包括过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、醌氧化还原酶1(NQO1)以及血红素加氧酶1(HO-1)。这些被激活的保护性酶类能够参与体内的广泛调节作用,对人体进行多方面的保护,研究发现该通路与机体炎症、肿瘤、衰老、凋亡、神经损伤、生育、妇产科疾病等多方面均有密切联系。综述Nrf2-ARE信号通路功能的研究进展,旨在全面客观地认识其生理功能。Nuclear factor E2 related factor 2(Nrf2) is a pivotal regulator of antioxidative stress. Under normal condition,its degradation is mediated in the cytoplasm and therefore don′ t exert physiologic function. When it was been activated by detrimental assault,Nrf2 can translocate into nuclear to bind with the antioxidant response element(ARE) then to form the Nrf2-ARE signal pathway,which subsequently activates a series of protective genes and proteins including catalase(CAT),superoxide dismutase(SOD),NAD(P)H:quinine oxidoreductase 1(NQO1) and heme oxygenase-1(HO-1). These enzymes play an important role in protecting somatic cells from detrimental assault. Researches have demonstrated that the pathway is associated with inflammation,tumor,aging,apoptosis,nervous injury and fertility as well as obstetrical and gynecological diseases closely. In order to understand the physiologic function of the pathway comprehensively,the present review is proposed.
