摘 要:目的观察参龟固本酒对大鼠酒精性肝损伤的作用。方法 SD大鼠按体质量随即分为3组:盐水对照组、白酒对照组及参龟固本酒组。各组分别灌胃给予生理盐水、相同酒精量的白酒、参龟固本酒液,每日2次,连续12周。末次给药后12h处死大鼠,采血,取肝组织,测定肝、脾指数;检测血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)活性及总蛋白(TP)、白蛋白(ALB)含量;检测血清及肝组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量;观察肝组织病理学改变。结果连续灌胃12周后,盐水对照组大鼠毛发、食量、粪便及活动等一般情况均无异常,体质量正常增长,各项生化指标均在正常生理值范围内。白酒对照组大鼠体质量增长缓慢,与盐水对照组比较,雄性大鼠体质量明显减轻(P<0.05),肝指数明显增高(P<0.01),脾指数无明显变化,血清ALT、AST、ALP活性较对照组均显著增加(P<0.01或P<0.05),血清SOD、MDA、TP、ALB无显著差异,而肝组织中SOD活性显著降低(P<0.05),MDA含量明显升高(P<0.05);同时,肝组织呈现大量大小不等的圆形脂滴空泡,可见点灶坏死及炎细胞浸润。参龟固本酒组大鼠体质量、肝、脾指数的变化明显轻于白酒对照组,与盐水对照组相比无明显差异(P>0.05);与白酒对照组比较,参龟固本酒组大鼠血清ALT、AST、ALP及肝匀浆MDA显著降低(P<0.01或P<0.05),而肝匀浆SOD显著增高(P<0.05);同时参龟固本酒组大鼠肝组织可见少量散在的脂滴空泡和炎细胞。结论参龟固本酒对大鼠肝损伤明显轻于同酒精量的白酒,其机制可能与药酒所含中药的抗氧化作用有关。Objective To observe the effect of Shen Gui Gu Ben Wine on alcoholic liver injury in rats.Methods SD rats were randomly divided into control group,white wine group and Shen Gui Gu Ben Wine group.Administration of different wine or normal saline twice a day began in rats by gastric gavage at the same time for 12 weeks.Samples were taken at the end of 12 th week.The followings were detected:liver and spleen index;activities of serum alanine aminotransferase(ALT),aspartate aminotransferase(AST)and alkaline phosphatase(ALP),the contents of total protein(TP)and albumin(ALB),superoxide dismutase(SOD)activity and malondialdehyde(MDA)content in serum and liver tissue;and the liver tissue pathology.Results After 12 weeks of continuous administration,rats'hair,appetite,faeces and physical activities in control group were unchanged.Compared with control group,rats in white wine group had significantly reduced weight(P〈0.05);significantly increased liver index(P〈0.01),increased serum ALT,AST,ALP and MDA contents in liver;and decreased SOD activity in liver;but there were no obvious changes in spleen index,the levels of serum TP,ALB,SOD or MDA(P 0.05).Pathological changes of liver tissue showed there were many fat drops,inflammatory cells infiltration and focal necrosis in white wine-treated rats.However,compared with white wine,Shen Gui Gu Ben Wine significantly increased rat body weight;decreased liver and spleen indexes,the levels of serum ALT,AST and ALP;enhanced liver SOD activities and decreased liver MDA contents(P〈0.01 or P〈0.05).Shen Gui Gu Ben Wine obviously alleviated hepatic steatosis and inflammation shown in white wine group.Conclusion Shen Gui Gu Ben Wine can improve alcoholic-induced liver injury in rats,the mechanism of which may be associated with the antioxidant compositions in Shen Gui Gu Ben Wine.
