摘 要:对皂角刺多糖的提取及抗氧化活性进行了研究。采用超声波辅助技术提取皂角刺中的多糖,用苯酚-硫酸法,以葡萄糖为标准对照物在波长484nm处测量吸光度,计算皂角刺中多糖含量。单因素试验和L9(34)正交试验得到的优化试验条件为:提取温度50℃、pH为7、提取时间40min、液料比为30比1。皂角刺中多糖的提取率为1.26%。抗氧化试验结果表明:当皂角刺多糖质量浓度为2.5g·L-1时,·OH、DPPH·及·O2-的清除率分别为88.9%,89.3%,42.3%。Extraction and antioxidation of polysaccharides in Chinese honeylocust spine was studied. The polysaccharide content of Chinese honeylocust spine was extracted with ultrasonic assisted technology and determined by phenol-sulfuric acid method using glucose as standard reference at the wavelength of 484 nm. The optimized conditions found by a series single-factor test and L9 (34) orthogonal experiment were as follows: extraction temperature was 50 ℃ ; pH was 7; extraction time was 40 min and ratio of liquid to solid was 30 : 1. Extraction yield of polysaccharide in Chinese honeylocust spine found was 1. 26%. The antioxidative experiments indicated that removal rates of · OH, DPPH · and ·O2- found were 88. 9%, 89. 3% and 42. 3%, respectively at mass concentration of polysaccharide in Chinese honeylocust spine of 2. 5 g · L-1.
