摘  要:目的研究D-阿洛糖对小鼠脑缺血再灌注损伤后血脑屏障的影响。方法 80只小鼠随机分为假手术组(sham组)、脑缺血模型组(MCAO组)、D-阿洛糖治疗组(D-allose组)、生理盐水对照组(NS组)。采用插线法制备小鼠大脑中动脉栓塞(MCAO)模型,尾静脉注射D-阿洛糖(300 mg/kg),运用改良小鼠神经功能缺损评分(m NSS)对小鼠脑缺血再灌注损伤程度评分;通过1%氯化三苯基四氮唑(TTC)染色,计算脑梗面积百分比;运用干-湿重法,测脑含水率;通过伊文思蓝(EB)含量反映血脑屏障的损伤程度;通过HE染色观察脑缺血再灌注损伤后梗死区的病理变化;采用免疫组化检测基质金属蛋白酶9(MMP-9)的表达。结果与MCAO组相比,D-allose可以明显降低小鼠脑缺血再灌注后神经功能缺损评分,减小小鼠脑梗死面积,减轻脑水肿,降低EB含量,减轻脑组织的病理损伤程度,并且明显减少MMP-9表达(P<0.05)。结论 D-阿洛糖可以保护小鼠脑缺血再灌注后的血脑屏障,其机制可能与抑制MMP-9的表达有关。Objective To study the effect of D-allose on on the blood-brain barrier permeability following focal cerebral ischemia/reperfusion in mice.Methods The focal cerebral ischemia-reperfusion model was induced by the middle cerebral artery occlusion(MCAO).80 male Balb/c mice were randomly divided into sham group, MCAO group, NS group, D-allose group.With a model of reversible middle cerebral artery occlusion in mice, D-allose was injected in caudal vein(300 mg/kg).The neurological deficit score was performed by modified neurological severity scores(mNSS), Infarct volume was calculated by triphenyl tetrazolium chloride (TTC) stain.Brain water content was measured by Dry/Wet method.Blood-brain barrier (BBB) permeability was evaluated by Evans blue extravasation. The histopathological changes was observed by HE staining and the expression of matrix metalloproteinase-9 (MMP-9) was detected by immunohistochemistry method.Results Compared with MCAO group, D-allose group significantly lowered neurological deficit score, reduced the infarct volume, alleviated brain water content markedly, D-allose diminished the extent and quantity of Evans blue extravasation(P〈0.05).Compared with MCAO group, D-allose decreased the level of MMP-9 expression (P〈0.05)and ameliorated the histopathological changes(P〈0.05). Conclusion D-allose has the protective effect on the blood-brain barrier following focal cerebral ischemia/reperfusion injury in mice, which is closely related to inhibition of MMP-9 expression.
