摘  要:目的探讨帕金森病(PD)前期多巴胺能神经元受损至出现局部病理改变前脑部葡萄糖代谢的改变。方法腹腔注射1-甲基-4-苯基-1,2,3,6四氢吡啶(MPTP)制作慢性PD小鼠模型,对照组注射生理盐水。于不同时间点,PET成像检测小鼠脑黑质及纹状体葡萄糖代谢的变化,蛋白电泳检测黑质酪氨酸羟化酶(TH)表达。结果小鼠双侧纹状体出现葡萄糖代谢降低时黑质致密部TH和磷酸化TH表达水平并无明显改变。结论PD小鼠模型双侧纹状体葡萄糖代谢降低早于黑质TH的下降。Objective To investigate the changes of brain glucose metabolism in mice at the early stage of Parkinson's disease (PD) when local pathological disorders does not appear. Methods A chronic PD model was established through intraperitoneal injection of MPTP into mice, while control mice were injected with normal saline. The changes of glucose metabolism in the substantia nigra and striatum were detected by PET imaging. The amounts of tyrosine hydroxy-lase (TH) and phosphor - TH ( p - TH) in the substantial nigra were measured by Western blotting. Results There was no remarkable change in the amounts of TH and p - TH in the substantial nigra when the glucose metabolism in the bilateral striatum of mice became weakened. Conclusion The weakening of glucose metabolism in the bilateral striatum appears before the levels of TH and p - TH are reduced in the substantial nigra.
