摘  要:目的探讨坎地沙坦对阿尔茨海默病(AD)大鼠认知功能的影响及机制。方法采用侧脑室注射β-淀粉样蛋白(AB)建立大鼠AD模型,利用Morris水迷宫检测坎地沙坦对大鼠的学习记忆的影响;通过免疫印迹方法检测大鼠海马区神经元型一氧化氮合酶(nNOS)表达水平的变化。结果Morris水迷宫结果显示给予坎地沙坦的AD模型大鼠找到平台的时间明显缩短,说明坎地沙坦可以明显改善AD大鼠的学习记忆能力;免疫印迹法显示坎地沙坦可以明显降低AB介导的nNOS磷酸化水平的升高。结论坎地沙坦可以改善AD大鼠认知功能,这可能与其降低Aβ介导的nNOS磷酸化水平的升高有关。Objective To investigate the effect of candesartan on the cognitive function using a rat model of Alzhei- mer's disease and potential mechanism. Methods A rat model of AD was established through stereotactical injection of β-amyloid (A13) into the bilateral ventricles of rats. The effects of candesartan on learning and memory were tested using the Morris Water Maze. The amounts of hippocampal nNOS was detected by Western blotting. Results According to the Morris Water Maze results, the rats receiving candesartan spent substantially less time to find the platform. It implied that candesartan can improve the learning and memory of AD rats. Candesartan treatment also significantly reduced an increase in the level of p - nNOS. Conclusion Candesartan can improve the cognitive function of AD rats, which may be related with its capacities to reduce the level of p - nNOS mediated by Aβ.
