摘 要:为给梵净山水菖蒲的合理开发和科学利用提供参考,采用乙醇加热回流提取法研究提取梵净山水菖蒲总黄酮的工艺条件,采用紫外分光光度法测定提取物对DPPH自由基和羟基自由基的清除率。结果表明:最佳提取工艺为乙醇浓度90%、提取2h、提取温度80℃、料液比1∶20。在该提取条件下,水菖蒲总黄酮的含量为40.83mg/g,提取液对DPPH自由基和羟基自由基具有一定的清除能力,当提取液总黄酮浓度为0.5mg/mL时,其DPPH自由基和羟基自由基的清除率分别达72.46%和73.48%。To provide the references for rational development and scientific utilization of A. calamus in Fanjing Mountain, the technological conditions of extracting total flavonoids from A. calamus in Fanjing Mountain were studied by ethanol refluxed back-flow, the antioxidant activity of the extract was evaluated by ultraviolet speetrophotometry on its ability of scavenging hydroxyl radical and DPPH radical. The results showed that the optimum procedure for ethanol refluxed back-flow of total flavonoids were ethanol concentration 90%, extracting time 2 h, extraction temperature 80℃ solid-liquid ratio 1 : 20, under the optimum conditions, the content was up to 40. 83 mg/g and the extracting solution has certain antioxidative effect. When the concentration of total flavonoid was 0. 5 mg/mL, the rate of scavenging DPPH radical and hydroxyl radical reached 72.46 G and 73.48 %.
