摘  要:目的优化红芪中芒柄花素和总皂苷的酶解提取工艺。方法选择复合酶用量、酶解时间、加水量和提取时间为试验因素,以芒柄花素及总皂苷提取量为指标,采用二次通用旋转组合设计优化红芪酶解提取工艺。结果提取芒柄花素的最佳工艺条件为复合酶用量340 mg,酶解时间110 min,加水量22倍,提取150 min;提取总皂苷的最佳工艺条件为复合酶用量280 mg,酶解时间90 min,加水量22倍,提取190 min。结论在上述优化条件下,两者提取量分别为69.99μg/g和1.68 mg/g。AIM To optimize the enzymatic extraction of formononetin and total saponin from Hedysari Radix.METHODS Complex enzyme concentration,enzymolysis time,water addition and extraction time were considered to be the influential experiment factors,the amounts of extracted formononetin and total saponin were taken into account as the evaluation indices,and the quadratic general rotary unitized design was applied to the enzymatic extraction process optimization. RESULTS The optimum conditions for formononetin were 340 mg enzyme concentration,110 min enzymolysis time,22 times of water addition and 150 min extraction time,while the best conditions for total saponin were 280 mg enzyme concentration,90 min enzymolysis time,22 times of water addition and 190 min extraction time. CONCLUSION In the aforementioned optimum conditions,the extraction volumes of formononetin and total saponin were 69. 99 μg / g and 1. 68 mg / g,respectively.
