摘 要:目的探讨胶质瘤原代培养细胞中基质金属蛋白活性酶-2(matrix metalloproteinase-2,MMP-2)对肿瘤细胞侵袭力的影响。方法对21例Ⅳ级胶质瘤组织进行原代培养并传代,通过明胶酶谱检测MMP-2酶原及活性酶含量,Boyden小室侵袭实验检测细胞侵袭力,分析MMP-2酶原及活性酶与肿瘤细胞侵袭能力之间的关系。结果 MMP-2活性酶的含量与肿瘤细胞侵袭力正相关(r>0,P<0.05),pro-MMP-2含量与肿瘤细胞侵袭力的相关性不具有统计学意义(P>0.05)。结论Ⅳ级胶质瘤中MMP-2活性酶是决定肿瘤细胞侵袭力的关键因素。Objective To study the influence of active MMP- 2 on invasive ability of Ⅳ grade glioma cells with primary culture. Methods The counts of all MMP- 2 forms in glioma cells by primary culture in 21 cases were detected by gelatin zymography,experiment of Boyden chamber were done to examine invasive ability,statistic analysis were made for the relationship between different forms of MMP- 2 with invasivness of glioma cells. Results There was positive relationship between the counts of active MMP- 2 with invasive ability of glioma cells( r 〉0,P 〈0. 05). The correlation analysis between pro MMP- 2 with glioma cells invasiveness got no statistical significance( P 〉0. 05). Conclusion Counts of active MMP- 2 form are very important factors to invasive ability of glioma cells.
