摘  要:目的探讨窖蛋白1(caveolin-1,Cav-1)在肺腺癌A549细胞凋亡中的作用及其分子机制。方法体外培养A549细胞,100 nM星形孢菌素(staurosporine,STS)处理6、12和24h后,流式细胞术检测STS处理后细胞周期的改变和凋亡率,Western blot方法检测STS诱导细胞凋亡过程中Cav-1、caspase-3、PI3K蛋白表达的变化。结果 FACS检测显示100 nM STS作用后,出现Ga/M阻滞和细胞凋亡,促凋亡作用于用药后6h出现,持续至24h,G_2/M阻滞的作用在12h达到峰值,凋亡率在24h达到峰值。Western blot结果显示经100 nM STS处理A549细胞6、12和24h后,Cav-1蛋白含量逐渐增加,组间比较差异均有统计学意义(P<0.01);caspase-3蛋白显示32kDa和17kDa条带,caspase-3含量逐渐增加,与对照组相比,STS处理12h和24h后,差异有统计学意义(P<0.01);P13K含量逐渐下降,与对照组相比,差异均有统计学意义(P<0.05)。采用caspase-3的抑制剂能阻止STS诱导的细胞凋亡。结论 Cav-1在STS诱导的A549细胞凋亡中起正调控作用。Objective This study was aimed to investigate carcinoma A549 cell line and its molecular mechanism. Methods the role of caveolin-1 in the cell apoptosis of lung adeno- A549 cells were cultured in vitro and 100 nm staurosporine (STS) bacteria treatment at 6h, 12h and 24h. Flow cytometry was used to detect alterations in cell cycle and apoptosis rate after STS treatment, and Cav-1, caspase-3 and PI3K proteins was detected by Western blot during STS induced cell apoptosis, Re- sults FACS assay showed that after 100 nm STS treatment, G2/M arrest and apoptosis could be detected; pro apoptotic role could be found at 6h after medication, continued to 24h; the role of G2/M arrest at 12h reached its peak, the apoptosis rate reached its peak at 24h. Western blot showed that after STS treatment A549 cells at 6h, 12h and 24h, Cav-1 protein content in- creased gradually, significant differences were observed among different groups (P〈0.01); caspase-3 protein showed 32kDa and 17kDa bands, caspase-3 content gradually increased, compared to the control group, the difference was significant after STS treatment at 12h and 24h(P〈0.01); PI3K content gradually decreased, compared with the control group, the differences were significant (P〈0.05). Caspase-3 can prevent cell apoptosis induced by STS. Conclusion Cav-1 plays a positive regulatory role in the apoptosis of A549 cells induced by STS.
