摘  要:[目的]探讨微小RNA-143(mi R-143)在结直肠癌组织中的表达及临床意义,以及上调mi R-143对于结直肠癌细胞凋亡的影响。[方法]采用实时荧光定量聚合酶链反应(real-time quantitative reverse transcription-PCR,real-time PCR)的方法在45例结直肠癌患者组织标本中检测mi R-143的表达水平,结合病理资料分析其临床意义。HCT-116细胞转染mi R-143 mimics后,用流式细胞术检测mi R-143对凋亡的影响。[结果 ]mi R-143在结肠癌组织中的相对表达量为0.48(0.24~0.91),在癌旁组织中的表达量为1.64(0.83~1.87),差异有统计学意义(P〈0.001),且在黏液癌患者中下调更明显(P〈0.05)。流式细胞术检测发现,与对照组的细胞凋亡率(20.00%±2.00%)相比,实验组的细胞凋亡率升高至31.83%±2.02%,差异有统计学意义(P=0.002)。[结论]mi R-143在结直肠癌组织中低表达,并且与组织学类型有关;mi R-143表达上调可促进HCT-116细胞凋亡;mi R-143具有潜在抑癌作用。[Purpose] To investigate the expression of mi RNA-143(mi R-143) in colorectal cancer and its significance,and to study the effect of up-regulation of mi R-143 on cell apoptosis of colonic cancer cell lines. [Methods] Real-time quantitative reverse transcription-PCR assay was applied to detect the expression level of mi R-143 in 45 samples of colorectal cancer tissues.Correlation between mi RNA expression levels and clinical characteristics of the patients were also analyzed. HCT-116 cells were transfected with mi R-143 mimics,and the cell apoptosis levels were detected by flow cytometry. [Results] The relative expression level of mi R-143 in colorectal cancer tissues was 0.48(0.24~0.91),which was significantly different compared with that in cancer adjacent tissues [1.64(0.83~1.87),P〈0.001]. The expression level of mi R-143 was significantly lower in mucinous carcinoma compared with that in adenocarcinoma(P〈0.05).The result of flow cytometry assay suggested that the ratio of apoptotic cells was(31.83% ±2.02%) in HCT-116 cells with overexpression of mi R-143(experimental group),which was significantly higher compared with that in control group [(20.00% ±2.00%),P =0.002]. [Conclusion] The expression level of mi R-143 is low in colorectal cancer tissue,and is related to the histologic type. Moreover,up-regulation of mi R-143 might promote the cell apoptosis of HCT-116 cells,which suggest mi R-143 has a potential role on inhibition of tumor.
