摘 要:目的研究左西孟旦体外抗氧化活性并考察其对培养心肌细胞缺氧性损伤的影响。方法采用1,1-二苯基苦基苯肼(DPPH)自由基、羟自由基及还原力3种体外抗氧化模型研究左西孟旦的抗氧化活力。建立大鼠乳鼠心肌细胞缺氧损伤模型,研究左西孟旦对缺氧培养心肌细胞的保护作用。结果质量浓度在0.05-0.5mg·mL^-1范围内,左西孟旦对羟自由基清除能力与维生素C组比,差异无统计学意义(P〉0.05);对DPPH·自由基清除能力弱于维生素C(P〈0.05);质量浓度在0.05-0.5mg·mL^-1范围内时,还原能力与维生素C差异无统计学意义(P〉0.05)。与模型组相比较,左西孟旦组能增强SOD的活性,降低MDA含量,提高清除自由基的能力(P〈0.01)。结论左西孟旦具有良好的体外抗氧化活性,对缺氧心肌细胞具有一定的保护作用。Objective To study in vitro the anti‐oxidative activity of levosimendan and protective effects of levosimendan on the cul‐tured hypoxic cardiomyocytes injury .Methods 3 in vitro models of anti‐oxidative activity were used to study ,including DPPH , hydroxyl free radicals and reducing power .Hypoxic injury model of cardiomyocytes in neonatal rats was established to investigate the protective effects of levosimendan .Results Within the range of 0 .05‐0 .5 mg · mL^ -1 ,there was no significant difference be‐tween levosimendan and vitamin C on scavenging capacities of hydroxyl free radicals (P〉0 .05) ,and scavenging capacities of levo‐simendan on DPPH · free radicals was weaker than that of vitamin C (P〈0 .05) .Within the range of 0 .05‐0 .5 mg · mL^ -1 ,there was no significant difference between levosimendan and vitamin C on reducing power (P〉0 .05) .Compared with the model group , levosimendan group enhanced the SOD activity ,reduced the production of MDA ,and protected the cultured hypoxic cardiomyo‐cytes injury from free radical(P〈0 .01) .Conclusion Levosimendan shows a good in vitro anti‐oxidative activity ,indicating levo‐simendan possesses good protective effects to hypoxic cardiomyocytes .
