摘  要:采用先进的统计学方法对桑树内生拮抗菌Bacillus subtilis X-16产抗真菌活性物质的发酵培养基进行了优化,以期得到最佳的培养基配方。以效价为指标,首先采用单次单因子试验筛选出培养基中的最佳缓效碳源、速效碳源、氮源,采用正交试验得出最佳的碳氮比(C/N)和无机盐,最后采用均匀设计试验。结果表明,获得最佳培养基配方:玉米粉10.0 g、蔗糖10.0 g、酵母浸膏粉5.0 g、(NH4)2SO43.6 g、Mg SO41.0 g、水1.0 L。经摇瓶发酵试验验证,发酵液效价较基础培养基提高了170.38%。X-16菌株发酵产生的抗菌物质对桑粘格胞菌表现出强烈的抑制作用,能够引起菌丝凝聚、破裂,导致原生质外溢。The fermentation medium of mulberry endophytic bacteria Bacillus subtilis X-16 that could produce antifungal active substance by advanced statistical designs was optimized to obtain the optimal medium. The titer of fermentation liquid was measured analytically. First,the slow release carbon source,the available carbon source,nitrogen source were screened out by one factor at a time,then the carbon: nitrogen ratio and inorganic salts were optimized by orthogonal testing,and then the uniform design experiment was used for the optimal media.The result showed that the optimized medium,including corn meal 10. 0 g,sucrose 10. 0 g,yeast extract powder 5. 0 g,( NH4)2SO43. 6 g,Mg SO41. 0 g,H2 O 1. 0 L,was good for the production of active substances. Production of antifungal active substance was170. 38 % higher than that of basal medium under optimum medium in shake flask fermentation,the crude antifungal active substances had a strong inhibition against Septogloeum mori L.,and the mycelium protoplast treated by antifungal active subtances was agglomerative and ruptured with protoplasm spillover.
