摘 要:采集拙政园3个底泥和3个土壤样品,运用末端限制性片段长度多态性(T-RFLP)技术分析拙政园6个样品的微生物多样性。研究结果显示:底泥细菌中T-RFs片段大小为181 bp、211 bp的菌种为优势菌种,土壤细菌中T-RFs片段大小为91 bp、205 bp的菌种为优势菌种。各样点底泥的真菌种类各不相同,且种类较少,无明显的优势菌种,而土壤真菌中T-RFs片段大小为355 bp的菌种为优势菌种。这说明拙政园底泥、土壤的细菌和真菌多样性存在差异,细菌多样性明显高于真菌。为苏州园林微生物多样性及群落结构累积了有价值的资料。Terminal restriction fragment length polymorphism (T-RFLP) technique was used to analyze microbial diversity in 3 points of sediment and 3 points of soil in Humble Administrator's Garden. The results show that among the sediment bacteria,T-RFs fragments with the sizes of 181 bp,211 bp were the dominant bacteria. A- mong soil bacteria,T-RFs fragments with the sizes of 91 bp, 205 bp were the dominant bacteria. Among sediment fungi, species of each site were not identical Among Soil fungi ,T-RFs fragment with the size of 355 bp was the dominant fungus. Bacterial and fungal diversity of sediment and soil in Humble Administrator's Garden were different. Bacterial diversity was significantly higher than that of fungi. This study has provided valuable data for microbial diversity and community structure of Suzhou gardens.
