摘 要:目的:探讨天然虾青素(ATX)对实验性大鼠肠缺血再灌注损伤的保护及作用机制。方法 :48只成年健康雄性SD大鼠随机分为假手术组(Sham组)、肠缺血再灌注组(IIR组)、高剂量ATX治疗组、低剂量ATX治疗组4组。Sham组仅分离肠系膜上动脉(SMA)而不夹闭;IIR组与ATX组于分离SMA后利用无损伤血管夹夹闭根部30 min、再灌注60 min诱导缺血再灌注损伤。两ATX治疗组分别于术前2 d给予ATX灌胃,剂量分别为100 mg/(kg·d)和50 mg(kg·d),连续灌胃2 d;Sham组及IIR组则均于同期给予相同剂量的生理盐水灌胃。4组大鼠于术后6 h处死,比较小肠组织丙二醛(MDA)、超氧化物歧化酶(SOD)、髓过氧化物酶(MPO)水平及血清中肿瘤坏死因子(TNF)-α、IL-1β、IL-6水平变化。结果:与Sham组及IIR组比较,两ATX治疗组小肠组织MDA水平、MPO活性及血清TNF-α、IL-1β、IL-6明显下降(P<0.05),而SOD活性则有明显上升(P<0.05),且高剂量组作用较低剂量组明显。结论:ATX可以减轻肠缺血再灌注损伤而起保护作用,其机制可能与抗氧化及抑制炎症反应相关。Objective:To explore the protective effect of Astaxanthin (ATX) on intestinal ischemia reperfusion injury (ⅡR) in rat,and find possible relationship to oxidative stress and inflammatory factors.Methods:36 SD rats were divided to four groups,as Sham group,ⅡR group,and two ATX treatment group (100 mg/kg and 50 mg/kg).The rats in two ATX group were dividely given 100mg/(kg· d) and 50mg/(kg· d) ATX solution(1 mL) by intragastric administration since 2 d before surgery,while other groups accept similar dose of saline.After 6 hours from operations,all rats were sacrificed,and liver MDA,SOD,MPO and serum TNF-α,IL-1 β,IL-6 were tested by ELISA kit.Results:The appliance of ATX gave rise to decreases in liver MDA,MPO and serum TNF-α,IL-1 β,IL-6,while SOD increased (P<0.05).The higher dose of ATX brought about more obvious effects.Conclusion:ATX can protect intestinie after ⅡR by anti-oxidation and inhibiting inflammation.
