摘  要:建立了高效液相色谱法同时测定苯丙氨酸解氨酶生物催化反应中L-苯丙氨酸和反式肉桂酸含量的方法。色谱分离选用Eclipse XDB-C18色谱柱(250 mm×4.6 mm,5μm);以1.5%乙酸和乙腈为流动相进行梯度洗脱,流速为0.8 m L/min;检测波长为260 nm。结果表明,该方法可在23 min内实现发酵液中L-苯丙氨酸和反式肉桂酸的完全分离与定量。该方法简便、快速、结果可靠,适合对L-苯丙氨酸和反式肉桂酸的定量检测。A high performance liquid chromatography (HPLC) method was developed to determine L-phenyalanine and trans-cinnamic acid in reaction catalyzed by phenylalanine ammonia-lyase. The chromatographic analysis was carried out by a Eclipse XDB-CI8 column (250mm×4.6 mm,5μm). The mobile phase was 1.5 % acetic acid and acetonitrile with gradient elution. The flow rate was maintained at 0.8 mL/min. The detection wavelength was set at 260 rim. Our results showed that this method could completely separate and quantify L-phenylalanine and trans-cinnamic acid within 23 rain. Thus, the proposed method is simple fast and reliable for the quantification of the L-phenyalanine and trans-cinnamic acid.
