摘 要:目的研究针对结缔组织生长因子(CTGF)的小干扰RNA(siRNA)对高糖诱导大鼠系膜细胞(RMC)合成血浆纤溶酶原激活物抑制剂-1(PAI-1)的抑制作用。方法取体外培养的RMC接种于含不同浓度的葡萄糖的6孔板中,设计并合成针对CTGF的siRNA(CTGF siRNA)并转染至高糖环境刺激的RMC中。利用逆转录-聚合酶链反应(RT-PCR)方法检测CTGF mRNA及PAI-1 mRNA在RMC中的表达,利用western blot法检测CTGF蛋白质及PAI-1蛋白质在RMC的表达。结果随着培养液中葡萄糖浓度的升高PAI-1 mRNA及PAI-1蛋白质在RMC中的表达明显增加,转染CTGFsiRNA后,明显减少。结论 CTGFsiRNA能明显抑制高糖诱导的RMC合成PAI-1。Objective To investigate the inhibitory effects of small interference RNA (siRNA) targeting connective tissue growth factor (CTGF) on plasminogen activator inhibitor 1 ( PAI-1 ) synthesized by high glucose-induced rat mesangial cell (RMC). Methods RMCs cultured in vitro were vaccinated in 6-well plates containing glucose of different concentrations ( 5 mmol/L, 10 retool/L, 15 mmol/L,20 mmol/L,25 mmol/L,25 mmol/L). The siRNA targeting CTGF ( CTGF siRNA) was designed and synthesized and then transfected into RMC stimulated by high glucose environment. The expressions of CTGF mRNA and PAI-1 mRNA in RMC were detected by reverse transcription polymerase chain reaction (RT-PCR) ,and the expressions of CTGF protein and PAI-1 protein in RMC were detected by western blot method. Results The expres- sions of PAI-1 mRNA 及 PAI-1 protein in RMC obviously increased with the increase of glucose concentration in culture fluid,while their expressions obviously decreased after transfeeted by CTGF mRNA. Conclusion CTGF siRNA could obvi- ously inhibit the synthesis of PAI-1 in high glucose-induced RMC.
