1型和2型糖尿病性白内障和正常晶状体的蛋白质组学分析

摘　　要：目的鉴定1型和2型糖尿病性白内障和正常晶状体内差异表达的蛋白质。方法通过饮食控制联合腹腔注射链脲佐菌素诱导1型和2型糖尿病大鼠模型,待典型白内障形成时,用强解聚试剂提取晶状体总蛋白,通过双向荧光差异凝胶电泳(two-dimensional fluorescence difference in-gel electrophoresis,2-D DIGE)分析,寻找差异蛋白质点。差异蛋白质点随后用激光辅助解吸/电离飞行时间质谱仪鉴定。结果 1型糖尿病性白内障(type 1 diabetic cataract,T1DC)的形成比2型糖尿病性白内障(T2DC)早,进展更迅速。2-D DIGE结果显示,与对照组相比,αA-、βA4-、βB2-晶状体蛋白在T1DC和T2DC晶状体内均显著减少;而βB1-、βA3-晶状体蛋白只在T2DC晶状体内明显减少;醛糖还原酶和γB-晶状体蛋白在T1DC晶状体内表达增多;而脂肪酸结合蛋白5、αB-晶状体蛋白在T2DC晶状体内表达增多。结论差异蛋白的不同可能是T1DC和T2DC临床表现和发病机制存在差异的原因。Objective To identify proteomic differences in type 1 （T1） and type 2 （T2） dia- betic cataracts （DCs） and normal lenses. Methods Type 1 and type 2 diabetes were induced by diet control and abdominal streptozotocin injection in rats. When characteristic cataracts were present in the diabetic rats, total proteins in the lenses were extracted using strong depoly- merizing reagents. Two-dimensional fluorescence difference in-gel eleetrophoresis （2-D DIGE） was carried out to detect differently-expressed protein spots. All differently-expressed protein spots were identified by matrix-assisted laser desorption/ionization time-of-flight （MALDI- TOF） mass spectrometry. Results Cataracts in type 1 diabetic rats were found earlier and progressed much more quickly than those in type 2 diabetic rats. αA-, βA4-, and βB2-crystal- lin were significantly decreased in both DC types compared with control group while βB1- and βA3-crystallin were only decreased in T2DC. Aldose reductase and γB-crystallin were marked- ly increased in T1DC while fatty acid-binding protein 5 and αB-crystallin were increased in T2DC lenses. Conclusion The differences in differently-expressed proteins might be the lead- ing cause of different clinical manifestations and pathogenesis.
