摘  要:目的:观察人染色体端粒酶基因(human telomerase RNA component,h TERC)和p27蛋白在宫颈病变中的表达情况,并探讨其在宫颈鳞癌发生发展过程中的作用及相互关系。方法:采用荧光原位杂交技术(florescence in situ hybridization,FISH)和免疫组化分别检测h TERC和p27蛋白在正常宫颈组织、宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)及宫颈鳞癌中的表达情况。结果:h TERC基因在对照组、CINⅠ、CINⅡ、CINⅢ及宫颈鳞癌组阳性表达率分别为5.0%(1/20)、22.2%(4/18)、72.7%(8/11)、81.8%(9/11)和96.7%(29/30),组间比较差异有统计学意义(P<0.05)。p27蛋白在对照组、CIN及宫颈鳞癌组阳性表达率分别为85%、35%、26.7%,组间比较差异有统计学意义(P<0.05)。双变量相关分析显示宫颈病变中,h TERC和p27蛋白表达呈负相关(r=-0.975,P=0.005)。结论:h TERC过度表达和p27蛋白表达水平下调均与宫颈鳞癌的发生发展相关,二者均可作为子宫颈癌变诊断及进展的预测指标。Objective: To explore the expression and relationship of telomerase h TERC with p27 proteins infection in cervical lesions. Methods: To investigate the expressions of telomerase h TERC and p27 proteins in cervical lesions and another 20 cases of normal cervical tissue were detected by FISH and immunohistochemically. Results: The positive expression rate of h TERC in normal cervical tissue,CINⅠ、CINⅡ、CINⅢ and cervical squamous cell carcinoma,was 5. 0%,22. 2%,72. 7%,81. 8% and 96. 7% respectively. The difference was statistically significant. The positive expression rate of p27 in normal cervical tissue,CIN,cervical squamous cell carcinomawas 85%,35% and 26. 7% respectively.The difference was statistically significant. According to bivariate correlation analysis,in cervical lesions,the positive expression rate of h TERC and p27 showed a significant negative correlation. Conclusion: h TERC overexpression and p27 protein down-regulated expression levels were associated with the development of cervical cancer. Both can be used as a predictor of cervical cancer diagnosis and progression.
