摘 要:目的:研究人肺癌干细胞的表面特征。方法:将肺癌细胞株制成单细胞悬液,接种到96孔板,每孔只有一个细胞,观察每孔细胞的存活,增殖以及分裂情况,将接种后能够持续分裂增殖的肿瘤细胞(实验组)和常规培养的肿瘤细胞(对照组)分别进行CD133细胞免疫组化,比较两组细胞的阳性表达率,将两组细胞按不同浓度梯度接种裸鼠,分析两组细胞的成瘤能力。结果:(1)仅有4.11%的肿瘤细胞具有增殖形成细胞克隆的能力。(2)单细胞体外培养具有增殖能力的细胞CD133阳性率明显高于常规培养的肿瘤细胞。(3)体外能够持续分裂的细胞成瘤能力强于常规培养的肿瘤细胞。结论:CD133可能是人肺癌干细胞的表面标志。Objective: To study the surface marker of lung cancer stem cell. Methods: Lung cancer cell were inoculated in96 shadow mask,and every hollow had only one cell. The survival,generation and cleavage of cells were observed. After inoculation,those tumor cells that could continue to split and proliferate( experimental group) and the conventional cultured tumor cells( control group) were undertaken CD133 immunohistochemistry respectively. The positive expression rates of the two groups were compared. The two groups of cells were inoculated to athymic mice according to different concentration gradient,and their ability of forming neoplasm was analyzed. Result:( 1) Only 4. 11% of the tumor cells has the ability of proliferation to form cell clones.( 2) Experiment group had higher positive expression rate of CD133 than control group.( 3) Forming neoplasm capability of experiment group was better than control group. Conclusion: CD133 may be the surface marker of lung cancer stem cell.
