摘  要:目的通过对全基因组启动子甲基化程度与视觉记忆强相关的基因进行生物信息学分析,寻找与记忆功能相关的基因所在信号通路。方法对9名正常人外周血单核细胞DNA样本进行甲基化DNA免疫共沉淀芯片分析。将基因的启动子区域甲基化水平与即刻及延迟视觉记忆功能进行相关性分析,筛选出与两者都强相关的基因并进一步进行通路和物理交互作用分析。结果与即刻及延迟视觉记忆都强相关的基因有69个,这些基因的通路及物理交互作用分析共涉及42条通路,其中22条通路的Q值小于0.05,包括了与能量代谢、突触功能、酪氨酸激酶活性、白细胞功能相关的通路。结论本研究首次利用甲基化组学数据与数量性状进行相关性及通路分析,揭示了可能与记忆相关并受甲基化调控的信号通路。Objective To explore the biological processes and pathways associated with memory function which may be regulated by gene promoter methylation. Methods The genome-wide promoter methylation statuses in 9 healthy individuals were analyzed with a Multiplex HG18 CpG Promoter chip. Genes with promoter methylation statuses strongly correlated with both immediate and delayed visual memory function were proceeded for pathway and physical interactions analysis. Results Sixty nine genes have been correlated with both immediate and delayed visual memory functions. Twenty two pathways, with a Q-value of 〈 0. 05, were identified by the pathway and physical interactions analysis, which included energy metabolism, axon guidance, tyrosine kinase activity, anterograde synaptic vesicle transport, and leukocyte migration and differentiation. Conclusion Pathways related with memory function may be regulated by DNA methylation.
