摘 要:目的探讨适宜浓度α-亚麻酸(ALA)对卵胞浆内单精子注射(ICSI)周期中人未成熟卵母细胞体外成熟、受精和胚胎发育的影响。方法选取2014年10月-2015年2月在该院接受ICSI治疗的不孕患者150名为研究对象,将其ICSI周期中未成熟卵母细胞随机分为ALA组(A组)、成熟卵泡液组(B组)和空白对照组(C组)。3组细胞在人未成熟卵母细胞体外成熟培养(IVM)培养液中培养24h,观察未成熟卵母细胞体外成熟情况及胚胎发育情况。同时测定卵母细胞培养基中超氧化物歧化酶(SOD)总活力和丙二醛(MDA)含量。结果A组24h成熟率高于B组,B组高于C组,差异均有统计学意义(P〈0.05);A组和B组卵母细胞ICSI后2PN卵裂率高于C组,差异有统计学意义(P〈0.05);3组2PN受精率、可利用胚胎率、囊胚形成率和SOD活力两两比较,差异无统计学意义(P〉0.05);A组MDA含量低于B组和C组,差异均有统计学意义(P〈0.05)。结论ALA通过降低氧化应激水平促进卵母细胞体外成熟,提高胚胎发育潜能。Objective To investigate the effect of appropriate dose of alpha-linolenic acid (ALA) on in vitro maturation (IVM), fertilization, and embryo development in intracytoplasmic sperm injection (ICSI) cycles. Methods From October 2014 to February 2015, a total of 150 infertile patients who received ICSI treatment were collected as study objects. Immature oocytes in ICS1 cycle were randomly di- vided into ALA group (group A), mature follicular fluid group (group B), and control group (group C ) . Cells in the three groups were cultured in IVM medium of human immature oocytes for 24 hours. The oocytes" maturation and embryo development were observed. Mean- while, superoxide dismutase (SOD) activity and malondialdehyde (MDA) levels were determined in the three groups. Results The 24- hour mature rate of immature oocytes in group A was significantly higher than that in group B ( P〈0. 05 ), and the rate in group B was higher than that in group C (P〈0. 05) , and the difference was statistically significant. In group A and group B, the cleavage rates of 2PN in oocytes after ICSI were higher than that in group C with statistical significance (P〈0.05) . In the three groups, rates of 2PN fertilization, available embryos and blastocyst formation, and SOD activity between either two groups revealed no statistical difference (P〉0. 05 ) . The level of MDA in group A was lower than those in group B and group C with significant difference (P〈0.05) . Conclusion ALA can promote IVM and increase embryo development potential in oocytes by reducing oxidative stress level.
